IMP3蛋白及Ki-67在脑胶质瘤中的表达及临床意义

摘　　要：目的探讨胰岛素样生长因子IImRNA结合蛋白3（IMP3蛋白）及Ki-67在脑胶质瘤中的表达以及临床意义。方法选取我院2008年1月至2012年10月手术切除的不同级别的胶质瘤60例（Ⅰ级5例,Ⅱ级20例,Ⅲ级20例,Ⅳ级15例）及5例正常脑组织,应用免疫组化Envision二步法检测IMP3蛋白及Ki-67在不同级别胶质瘤中的表达,分析二者与胶质瘤恶性程度的关系以及二者的相关性。结果 IMP3在胶质瘤各级别的表达中有统计学意义（χ2=41.17,P〈0.05）,其中胶质瘤Ⅲ~Ⅳ级阳性表达率明显高于Ⅰ~Ⅱ级（91.4%vs8%,χ2=41.34,P〈0.05）。胶质瘤标本的Ki-67指数（Ki-67LI）范围为0%~50%,在胶质瘤各级别的表达中有统计学意义（χ2=29.80,P〈0.05）,其中胶质瘤Ⅲ~Ⅳ级阳性表达率明显高于Ⅰ~Ⅱ级（91.4%vs 20%,χ2=50.29,P〈0.05）。IMP3蛋白的阳性表达率与Ki-67蛋白的阳性表达率有正相关性（r=0.774,P〈0.05）。结论 IMP3及Ki-67在脑胶质瘤中的表达均与胶质瘤的恶性程度呈正相关,二者联合应用可作为评价胶质瘤级别的重要参考指标。Objective To investigate the expression and clinical significance of insulin-like growth factor II mRNA-binding protein 3（IMP3）and Ki-67 in human brain glioma.Methods 60 different grade of gliomas were selected from2005 to 2012 in our hospital（grade I 5 cases,grade II 20 cases,grade III 20 cases,grade IV15 cases）and 5 cases of normal brain tissue.The expression of IMP3 and Ki-67 was detemined in gliomas using immunohistochemistry（Envision method）..The correlation was analyzed between expression of IMP3,Ki-67 and malignant degree of gliomas.And the correlation between the expression of IMP3 and Ki-67 was also analyzed.Results The expression of IMP3 in different grade gliomas was statistically significant（χ2=41.17,P 0.05）and it was significantly higher in gradeⅢ-Ⅳ gliomas than that in gradeⅠ-Ⅱ（91.4% vs.8%,χ2=41.34,P0.05）.The range of Ki-67 LI was 0%-50%.The expression of Ki-67 in different grade gliomas was statistically significant（χ2=29.80,P0.05）and it was significantly higher in grade Ⅲ-Ⅳ gliomas than that in gradeⅠ-Ⅱ（91.4% vs.20%,χ2=50.29,P0.05）.There was positive correlation between IMP3 and Ki-67 expression in brain gliomas（r=0.774,P0.05）.Conclusion The expression of IMP3 and Ki-67 in gliomas was positive correlated with the grades of gliomas.The united application of them can be used as important reference index for evaluation of gliomas＇ s biological behavior.
