摘  要:目的观察藤茶提取物对2型糖尿病(T2DM)大鼠胰岛素抵抗的改善作用及可能的分子机制。方法雄性SD大鼠高脂喂养联合小剂量链脲佐菌素腹腔注射(STZ,30 mg/kg)建立T2DM大鼠模型。建模成功后将T2DM大鼠系统抽样随机分为糖尿病模型对照组(DIA)、二甲双胍干预组(MET,125 mg/kg)、二氢杨梅素干预组(DHM,100 mg/kg)、藤茶提取物低剂量干预组(R50,50 mg/kg)、中剂量干预组(R100,100 mg/kg)及高剂量干预组(R200,200 mg/kg)6组。每组大鼠分别灌胃给药干预4周后,放射免疫分析法测定大鼠血清中胰岛素、胰岛素C肽;Western blot法检测肝脏组织中p-Akt、FGF21、p-AMPK蛋白表达;qRT-PCR法检测肝脏组织中FGF21 mRNA的表达。结果与DIA组相比,二甲双胍、DHM和藤茶提取物干预组大鼠血糖、血清胰岛素水平、HOMA-IR指数显著降低(P<0.05),而胰岛素C肽水平显著升高(P<0.05);同时肝脏组织中p-Akt、FGF21、p-AMPK蛋白及FGF21 mRNA的表达升高(P<0.05)。结论藤茶提取物可通过上调p-Akt、FGF21、p-AMPK的表达从而改善2型糖尿病大鼠的胰岛素抵抗。Objective To determine the effects of Rattan tea on insulin resistance in rats of type 2diabetes( T2DM) and investigate the possible underlying molecular mechanism. Methods Male SD rats( SPF) fed with high-fat diet and injected with small dose of streptozotocin( STZ) were used to establish type2 diabetic rat model. Then,T2 DM rats were randomly divided into diabetic model group( DIA),metformin125 mg / kg group( MET),dihydromyricetin 100 mg / kg group( DHM),Rattan tea extracts 50 mg / kg group( R50),Rattan tea extracts 100 mg /kg group( R100) and Rattan tea extracts 200 mg /kg group( R200).After 4 weeks' intervention,the serum levels of insulin and insulin C-peptide,and the protein expression of pAkt,FGF21 and p-AMPK were detected by Western blotting,as well as the mRNA expression of FGF21 in liver tissues were measured by real-time RT-PCR. Results Compared with DIA group,the levels of blood glucose,insulin and HOMA-IR were decreased significantly,meanwhile,the serum level of insulin C-peptide was increased significantly,the protein expression of p-Akt,FGF21,p-AMPK and FGF21 was up-regulated in liver tissues,as well as mRNA expression of FGF21( P〈0. 05). Conclusion Rattan tea improves insulin resistance in diabetic rats via up-regulating the expression of p-Akt,FGF21 and p-AMPK.
