摘  要:目的:探索性地研究一种非凋亡性死亡方式—自噬是否为青蒿琥酯(ART)诱导胰腺癌细胞死亡的途径,为青蒿琥酯治疗胰腺癌提供实验依据,为探寻治疗胰腺癌的新策略提供思路。方法 :经丫啶橙(AO)染色、荧光显微镜及流式细胞仪检测细胞内自噬溶酶体;并应用自噬抑制剂3-甲基腺嘌呤(3-MA)观察抑制自噬对ART介导的Panc-1细胞存活率及细胞周期的影响。结果:经丫啶橙染色后提示阴性对照组和ART处理组细胞均存在自噬,两者无明显差异(P>0.05);流式细胞仪检测显示ART作用于胰腺癌Panc-1细胞并没有诱导细胞内自噬溶酶体明显增加,应用自噬特异性抑制剂3-MA预处理后,荧光显微镜显示抑制自噬对细胞的生长状态无明显影响,台盼蓝排斥实验表明,抑制自噬对ART介导的Panc-1细胞存活率无明显影响;流式细胞仪检测结果显示抑制自噬对ART介导的Panc-1细胞G2/M期阻滞无明显影响(P>0.05)。结论:ART诱导胰腺癌Panc-1细胞死亡的过程中存在自噬,但自噬并非ART诱导产生,也不是引起胰腺癌细胞死亡的方式。Objective:To explore the role of autophagh in the death way of pancreatic cancer cells induced by artesunate.Methods:The autophagy lysosomal of intracellular was detected by acridine orange(AO)staining,fluorescence microscopy and flow cytometry;and autophagy inhibitor 3-methyl adenine(3-MA)was used to inhibit autophagy.ART-mediated survival rate of Panc-1cells and the change of cell cycle was also observed.Results:By acridine orange(AO)staining,fluorescence microscopy showed that there was no significant difference in autophagic cells between negative control group and ARTtreatment group(P〉0.05);flow cytometry showed that there was not significant increase of autophagy lysosomal in the ARTtreatment group.After specific autophagy inhibitor 3-MA pre-treatment,the fluorescence microscope showed that the cell growth state had no significant impact after autophagy inhibition.Trypan blue exclusion experiments showed that after the inhibition of autophagy,there was no significant change in survival rate of Panc-1cells in the ART-treatment group.Flow cytometry results showed that after the inhibition of autophagy,there was no significant change in Panc-1cells in G2/ M phase arrest in the ART-treatment group(P〈0.05).Conclusions:Autophagy exists in the death process of ART-induced pancreatic cancer Panc-1cell,but it is neither ART-induced,nor is the way caused pancreatic cancer cell death.
