声动力疗法刺激肿瘤微环境中CD80与CD86表达的研究

摘  要:[目的]检测声动力治疗(sonodynamic therapy,SDT)对B16F10荷瘤鼠模型中肿瘤微环境T细胞共刺激分子CD80、CD86表达情况的影响。[方法]以2.0W/cm2超声强度结合250mg/kg 5-氨基酮戊酸(5-ALA)治疗B16F10荷瘤鼠,测量不同实验组小鼠体重及肿瘤大小;免疫组织化学方法检测移植瘤微环境中CD80、CD86、IFN-γ和TNF-α表达情况。[结果]与其他组相比,SDT组肿瘤体积缩小更加显著,体重变化无差异;SDT组CD80、CD86、IFN-γ和TNF-α均出现表达增高,且具有统计学意义。[结论]SDT能够加强T细胞共刺激分子的CD80、CD86的表达,促进抗肿瘤免疫的发生。[Purpose] To detect the variation of costimulatory molecules CD80 and CD86 in tumor microenvironment by sonodynamic therapy (SDT) in B16FI0 tumor-bearing nude mice. [Methods] 2W/cm^2 ultrasound intensity with 250mg/kg 5-Aminolevulinic acid (5-ALA)were used in B16F10 tumor-bearing nude mice,and the tumor volume and body weight of mice were measured in different groups respectively before and after treatment.CD80,CD86,IFN-γ and TNF-α in tumor microenvironment were detected by immunohistochemistry. [Results] Compared with other groups, variation of tumor volume in SDT group was statistically significant, while vari- ation of body weight in SDT group was not significant; and expressions of CD80, CD86, IFN-γand TNF-α in SDT group increased significantly. [Conclusion ] SDT facilitates costimulatory molecules CD80 and CD86 secretion and promotes anti-tumor immunity.
