摘 要:[目的]探讨成纤维细胞生长因子诱导14(Fn14)与表皮生长因子受体(EGFR)外显子19缺失突变之间的关系。[方法]对125例EGFR外显子19缺失突变的非小细胞肺癌(NSCLC)患者,应用免疫组织化学法检测组织样本中EGFR 19缺失(EGFR 19-Del)和Fn14的表达,统计分析EGFR 19-Del与Fn14在不同临床病理特征中的表达差异以及两者之间的相关性。[结果]EGFR 19-Del阳性主要在细胞膜上表达,Fn14阳性表达主要定位于胞膜和胞质中。EGFR 19-Del的阳性表达率为99.2%,Fn14阳性表达率为100.0%。EGFR 19-Del、Fn14在鳞癌、腺癌组间的表达差异均无统计学意义(P=0.480;P=0.106);EGFR 19-Del、Fn14在总的NSCLC、腺癌的不同病理分级、TNM分期中差异显著(P均<0.05),而在鳞癌的不同病理分级、TNM分期中差异无统计学意义(P均>0.05)。Fn14与EGFR 19-Del在总的NSCLC、腺癌、鳞癌中的表达均呈正相关(P均<0.05,rs均>0.47);二者在中、低分化癌中呈正相关(P均<0.05,rs均>0.43),而在高分化癌中无相关性(rs<0.3);二者在Ⅱ、Ⅲ期中呈正相关。[结论 ]Fn14在NSCLC EGFR 19缺失突变样本中有很高的表达率,其表达与EGFR 19-Del有相关性,提示EGFR激活突变可能会影响Fn14的表达。[ Purpose] To investigate the correlation of fibroblast growth factor-inducible 14 (Fn14) expression with epidermal growth factor receptor (EGFR) exon 19 deletion mutation. [Methods] Fn14 and EGFR 19-Del were detected in 125 NSCLC samples with EGFR exon 19 deletion mutation immunohistochemically. [ Results ] The expression of EGFR 19-Del was mainly located in cell membrane and the expression of Fn14 was mainly located in cell membrane and cytoplasm. The expression rates of EGFR 19-Del and Fn14 proteins were 99.2% and 100.0%,respectively. There was no significant differences of EGFR 19-Del and Fn14 between squamous cell carcinoma group and adenocarcinoma group(P=0.480; P=0.106). There were significant differences of EGFR 19-Del, Fn14 among different pathological grades and different TNM stages in NSCLC or adenocarcinoma (P〈0.05), while there was no statistical difference in squamous cell carcinoma(P〉0.05). The correlation of Fn14 with EGFR 19-Del were positive (P〈0.05,rs〉0.47) in NSCLC,adenocarcinoma or squamous cell carcinoma. The correlation of Fn14 with EGFR 19-Del were positive in middle or low differentiation patients (P〈0.05, rs〉0.43), but no relevance was found in high differentiation of NSCLC (rs〈0.3). And there were positive correlations between Fn14 and EGFR 19-Del in patients with stage II or III. [Conclusion] The Fn14 protein is highly expressed in NSCLC with EGFR exon 19 deletion mutation,and its expression has correlation with the expression of EGFR 19-Del, which suggests that EGFR activated mutation might influence the expression of Fn14 .
