摘 要:目的研究氯化钴对胶质瘤T98G细胞上皮间充质转化(EMT)和侵袭能力的影响。方法用氯化钴处理胶质瘤T98G细胞,未处理组作为对照,Western blot检测各组细胞上皮细胞钙黏蛋白(E-cadherin)和间充质细胞波形蛋白(Vimentin)的表达,采用Transwell及Boyden方法检测各组迁移细胞数。结果氯化钴处理胶质瘤T98G细胞后发生了典型的EMT:E-cadherin表达明显降低(P<0.05或P<0.01),Vimentin表达明显升高(P<0.01);Transwell迁移实验显示迁移能力明显增强:穿过Transwell小室滤膜的细胞数由(29±5)个增加至(53±4)个(P=0.0397);Boyden结果显示:穿过小室细胞数由(26±3)个增加至(36±2)个(P=0.0402)。结论氯化钴能促进胶质瘤T98G细胞发生EMT,增强胶质瘤T98G细胞迁移能力。Objective To investigate the effect of CoCl2 on epithelial-mesenchymal transition(EMT) and the invasion ability of glioma T98G cells. Methods The human glioma T98G cell was induced by CoCl2, those without any treatment was taken as control group. The expression of epithelial cell E-cadherin and mesenchymal cell vimentin in all groups was detected by Western blot. Moreover, the cell invasion number in all groups was examined by Transwell and Boyden chamber assays. Results After glioma T98G cell was induced by CoCl2, typical EMT was occurred: the expression of Ecadherin was decreased significantly(P〈0.05 or P〈0.01), the expression of vimentin was increased significantly(P〈0.01); Transwell migration assay showed that the transfer ability was increased significantly: the cell number of passing through filter membrane of Transwell cella increased from(29 ±5) to(53 ±4)(P = 0.0397); Boyden results showed that the cell number of passing through cella increased from(26±3) to(36±2)(P = 0.0402). Conclusion CoCl2 can promote EMT of glioma T98G cells, and increase the invasion ability of glioma T98G cells.
