摘 要:目的探讨低频低能量超声联合微泡抑制人前列腺癌细胞转移及分子机制。方法以频率21 k Hz、声强46 m W/cm2超声,采用连续波辐照人前列腺癌细胞PC-3悬液30 s。分为三组:对照组、单独超声辐照组及超声辐照联合微泡组(加声诺维200μl)。辐照后培养12 h,Transwell小室模型进行细胞体外转移实验,western-blot检测其基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)表达。结果辐照后12 h,与对照组比较,单独超声辐照组及超声辐照联合微泡组细胞转移数量受到抑制,并且以超声辐照联合微泡组受到抑制最明显(均P<0.05);细胞辐照24 h后,与对照组比较,单独超声辐照组及超声辐照联合微泡组MMP-2及MMP-9蛋白表达明显受到抑制,并且以超声辐照联合微泡组最为明显(均P<0.05)。结论低频超声联合微泡能够抑制人前列腺癌细胞PC-3的转移,并且可能通过下调MMP-2、MMP-9蛋白表达来实现。Objective To study whether ultrasound combined with microbubbles could suppress the metastasis of androgen-independent prostate cancer cells PC-3 and to identify the probable mechanism. Methods Androgen-independent human prostate cancer PC-3 cells were irradiated continuously by ultrasound with the frequency of 21 k Hz and intensity of46 m W/cm2 for 30 s. The PC-3 cells were divided into three groups: the control group, the ultrasound group and the ultrasound combined with microbubbles(200 μL Sono Vue) group. Twelve h after treatment,transwell chamber model was applied to do cell metastasis experiments in vitro. Western-blot was used to measure the expression of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-2(MMP-9) protein. Results Twelve h after treatment, ultrasound combined with microbubbles could significantly inhibit the metastasis of human prostate cancer cells(P 0.05). The expression of MMP-2 and MMP-9protein was downregulated by ultrasound combined with microbubbles(P 0.05). Conclusion Ultrasound combined with microbubbles could inhibit the metastasis of human androgen-independent prostate cancer cells through down-regulation of MMP-2 and MMP-9 expression.
