摘 要:目的通过观察大鼠胰腺缺血模型中红景天苷的作用,探讨红景天苷对胰腺炎缺血改变的机制。方法 200只Wistar大鼠随机分为3组,假手术组(50只)、对照组(75只)及试验组(75只),通过血管阻断方法建立胰腺缺血模型。假手术组行开腹关腹手术,对照组建立大鼠胰腺急性缺血模型,试验组建立缺血模型后给予静脉注射红景天苷。检测各组血淀粉酶、血栓烷素B2(TXB2)、白细胞介素8(IL-8)的水平。结果大鼠缺血模型建成后,试验组4 h血淀粉酶水平开始下降,在8 h及12 h时点与对照组对比,差异有统计学意义(P<0.05);在8 h及12 h时点,试验组TXB2水平明显低于对照组,差异有统计学意义(P<0.05);对照组12 h内TXB2水平呈现持续升高趋势,试验组4 h后TXB2呈现下降趋势,与对照组对比,在12 h时点差异有统计学意义;对照组12 h内IL-8水平持续升高,试验组在12 h时点开始下降,与对照组对比,差异有统计学意义(P<0.05)。结论中药红景天苷对大鼠ANP缺血损伤有明显改善作用,能够降低炎性因子及胰酶水平。Objective To observe the effect of salidroside on ischemia in rats with acute necrotizing pancreatitis(ANP) model and discuss the mechanism.Methods Two hundred Wistar rats were divided into three groups randomly:sham operation group(n =50) control group(n =75) and experiment group(n =75).Rats in control group and experiment group were made to acute necrotic pancreatitis model by artery blocking and the sham operation group were treated by abdominal incision,and the rats in experiment group were treated with salidroside i.v.Blood sample was drawn at 0,2nd,4th,8th,12 th h after reperfusion,and the levels of blood amylase,TXB_2,IL-8 were detected.Results At the 4th h,blood amylase began to decrease in experiment group,compared with control group at the8 th and 12 th h,the differences were significant(P 0.05);At 8th h and 12 th h,TXB_2 in experiment group was significantly lower than that in control group(P 0.05);TXB-2 in control group increased at 12 th h and TXB_2 in experimental group began to decrease at 4th h,compared with control group,the difference was significant at the 8th h(P 0.05);IL-8 in control group increased in 12 h and IL-8 began to decrease at 12 th h,compared with control group IL-8 decreased significantly at the 12 th h(P 0.05).Conclusion Salidroside could protect ischemia injury in rats with ANP and decrease the levels of amylase,TXB_2 and IL-8.
