BRCA1 BRCA2在结直肠息肉和癌中的表达及临床意义

摘　　要：目的:检测同源修复基因家族成员BRCA1、BRCA2在结直肠息肉和癌中的表达,探讨其与临床病理特征的关系。方法:利用免疫组化SP法分别检测27例正常结直肠粘膜、30例结直肠增生性息肉、33例结直肠腺瘤性息肉、60例结直肠腺癌组织中BRCA1、BRCA2的表达水平。结果:BRCA1、BRCA2在正常结直肠粘膜、结直肠增生性息肉、结直肠腺瘤性息肉、结直肠癌组织中的表达率逐渐增高,差异有显著性(P<0.05),二者的表达与结直肠癌中的性别、年龄、分化程度、Ducks分期等均无统计学差异(P>0.05);二者在结直肠癌组织中的表达水平呈正相关(r=0.295,P<0.05)。结论:BRCA1、BRCA2表达呈正相关;二者可能参与结直肠癌的发生、发展;联合检测结直肠病变组织中的BRCA1蛋白、BRCA2蛋白的表达水平有助于大肠癌的早期发现。Objective： To detect members of the family of homologous repair genes BRCA1 and BRCA2 expression in colorectal polyps and cancer,to explore its relationship with the clinical pathological features.Method： By using immunohistochemical SP method detected 27 cases of normal colorectal mucosa,30 cases of colorectal hyperplastic polyp,33 cases of colorectal adenomas sex polyp,60 cases of colorectal adenocarcinoma tissue protein BRCA1 and BRCA2 protein expression,analysed the physical index and the correlation with colorectal polyps and cancer. Result： BRCA1 and BRCA2 in normal colorectal mucosa,colorectal hyperplastic polyp,colorectal adenomas polyps and colorectal cancer expression rate increased gradually in the organization,and had significant difference（ P〈0. 05）,the expression in colorectal cancer such as sex,age,degree of differentiation,Ducks stage had no statistical difference（ P〉0. 05）,both in the tissue of colorectal cancer expression level significantly positively correlated（ r = 0. 295,P〈0. 05）. Conclusion：Expression of BRCA1 and BRCA2 are positively correlated,both may participate in the occurrence and development of colorectal polyps and cancer. Joint detection of colorectal lesions in the organization the expression of BRCA1 protein and BRCA2 protein levels for early detection of colorectal cancer.
