摘  要:目的:探讨Cullin1(Cul1)基因对脑胶质瘤细胞U251细胞周期的影响。方法运用Control siRNA、Cul1 siRNA分别转染脑胶质瘤细胞U251,Western blot检测Cul1蛋白表达,流式细胞仪检测脑胶质瘤细胞U251细胞周期的变化,Western Blot 检测Cul1基因干涉后对脑胶质瘤细胞U251细胞周期蛋白及细胞周期蛋白依赖性激酶抑制剂的影响。结果与Control siRNA组相比,Cul1 siRNA组脑胶质瘤细胞U251 Cul1蛋白表达量降低99.1%;Cul1基因干涉后脑胶质瘤细胞U251细胞G1期的细胞比例上升比较明显;Cul1基因干涉后脑胶质瘤细胞U251中的Cyclin D1及Cyclin E的蛋白表达量明显减少,而p21和p27的蛋白表达量明显增加。结论 Cul1 siR-NA靶向干扰了Cul1的表达,通过下调Cyclin D1、Cyclin E蛋白的表达,同时上调p21和p27蛋白的表达,使脑胶质瘤细胞U251停滞在G 1期。Objective To investigate the effects of Cullin1 (Cul1) on the cell cycle of glioma U251 cells.Methods The expression of Cul1 in glioma U251 cells was silenced through transfection of Cul 1 siRNA.Then the presence of Cul1 protein was detected using Western blotting. Changes in the cell cycle of U251 cells were measured by flow cytome-try.Meanwhile , the expression of cell cycle proteins and cyclin -dependent kinase inhibitors were examined after Cul 1 silencing .R seults Compared with the control , Cul1 siRNA could knock down the expression of Cul 1 by 99.1%in glio-ma U251 cells and remarkably increase the percent of cells in G 1 phrase.After Cul1 silencing, the expression of cyclin D1 and E proteins were drastically reduced , while the levels of p21 and p27 protein were significantly enhanced .Conclu-sion Knockdown of Cul 1 can cause cell cycle arrest at G 1 phrase through down-regulating the expression of cyclin D 1 and E, in combination with up-regulating the levels of p21 and p27.
