摘 要:目的研究雌激素受体β(ERβ)对阿尔茨海默病大鼠海马内炎症反应及淀粉样β蛋白沉积的影响,并探讨其可能的机制。方法β淀粉样蛋白脑室内注射建立阿尔茨海默病大鼠模型,并将模型分为对照组、ERβ组和shRNA组。对照组不做任何处理,ERβ组和shRNA组大鼠分别通过脑室内注射载有雌激素受体β基因或shRNA的慢病毒质粒。通过Morris水迷宫检测各组大鼠行为学,Western blot检测各组大鼠脑组织内Akt及β淀粉样蛋白表达水平,qRT-PCR检测TNF-α和IL-1βmRNA表达,并通过免疫荧光染色检测Aβ的表达。结果ERβ组大鼠记忆能力明显高于对照组和shRNA组大鼠(P<0.05),ERβ组大鼠海马内Akt含量明显高于对照组和shRNA组大鼠(P<0.05),并且ERβ组大鼠海马内IL-1和TNF-αmRNA以及β淀粉样蛋白含量明显低于对照组和shRNA组大鼠(P<0.05)。结论过表达ERβ在不依赖雌激素的情况下可减轻AD大鼠海马内炎症反应和β淀粉样蛋白的沉积,改善AD大鼠行为学的变化,其神经保护作用可能是通过激活Akt途径实现。Objective To study the influence of estrogen receptor beta on inflammation and the amyloid beta deposition in hippocampal of alzheimer's disease rat model,and investigate its possible mechanism. Methods Amyloid beta was injected into ventricle to establish the rat model of alzheimer's disease,and all models were divided into control group,ERβand shRNA groups. Control group was without any treatment,ERβ and shRNA group rats were injected with ER beta plasmid and ER beta shRNA lentivirus plasmid respectively. Ethology of each rat was determined by Morris water maze test.Akt and Amyloid beta rat brain tissue were detected using Western blot. qRT-PCR was utilized to detect TNF-alpha and IL-1 beta mRNA expression,and expression of Amyloid beta was diplayed by immunofluorescence staining. Results The memory ability of rats ER beta group was obviously higher than that of control group and the shRNA group rats( P〈0. 05).ER rat hippocampal Akt content in Eβ group was obviously higher than that of control group and the shRNA group rats( P〈0. 05). mRNA expression of TNF-alpha and IL-1β in the rat hippocampus of ER beta groups,as well as Amyloid beta content were significantly lower than the control group and shRNA group rats( P〈0. 05). Conclusion Overexpression of ER beta could reduce the AD rat hippocampal inflammation and A beta deposition,behavioral deficits in the rat model of AD,and the neuroprotective effect may be achieved by activation of Akt pathway.
