摘  要:目的:观察莫诺苷对局灶性脑缺血-再灌注大鼠细胞周期蛋白Cyclin D1及CDK6的影响。方法:用线栓法制备大鼠局灶性脑缺血-再灌注模型。将15只Sprague-Dawley(SD)大鼠随机分为假手术组,模型组,莫诺苷组(270、90、30 mg·kg-1)。利用免疫组化方法检测大鼠患侧海马Cyclin D1、CDK6表达。结果:与假手术组相比,模型组Cyclin D1及CDK6表达显著增加;给予莫诺苷治疗后,与模型组相比,莫诺苷(270、90 mg·kg-1)剂量组能显著降低大鼠Cyclin D1及CDK6的表达。结论:莫诺苷能通过降低脑缺血-再灌注后Cyclin D1及CDK6的表达而发挥神经保护作用。Objective: To explore the effects of morroniside on key cell-cycle regulator proteins Cyclin D1 and CDK6 in a rat model of focal cerebral ischemia-reperfusion(I-R). Methods: After the model of middle cerebral artery occlusion(MCAO) was established,15 Sprague-Dawley(SD) rats were randomly assigned into sham group,model group,morroniside group(270、90、30 mg·kg-1). Immunohistochemistry staining was used to measure the expression of Cyclin D1 and CDK6 in ischemic ipsilateral hippocampus. Results: Compared with sham group,the expression of Cyclin D1 and CDK6 were signifi cantly increased in vehicle-treated I-R rats. However,the expression of Cyclin D1 and CDK6 was signifi cantly reduced by treatment of morroniside at doses of 270 mg·kg-1and 90 mg·kg-1 after I-R compared with control group. Conclusion: Morroniside might play a role of neuroprotection by down-regulating the expression of Cyclin D1 and CDK6 after cerebral I-R treatment.
