摘　　要：目的：探讨前列腺素转运蛋白（PGT）在结直肠癌组织中的表达及其与临床病理特征的关系。方法采用实时荧光定量PCR技术、Western blot和免疫组织化学（免疫组化）技术检测80例结直肠癌组织和癌旁组织标本中PGT mRNA和蛋白的表达，并分析其与临床病理特征的关系。结果与结直肠癌旁正常组织相比，癌组织中PGT mRNA相对表达量[（0．57±0．33）比（2．33±1．20）]、蛋白表达水平[PGT／甘油醛-3-磷酸脱氢酶（GAPDH）灰度值（0．45±0．16）比（0．78±0．23）]、免疫组化累计吸光度值[（718．7±359．4）比（10412．0±6423．3）]和平均吸光度值[（0．03±0．01）比（0．12±0．09）]均明显降低（均P＜0．01）。结直肠癌患者癌组织中mRNA和蛋白的低表达与肿瘤浸润深度（T3～T4）和分期晚（Ⅲ～Ⅳ期）有关（均P＜0．01）；而与性别、年龄、肿瘤位置及分化程度无关（均P＞0．05）。结论结直肠癌组织中PGT mRNA和蛋白的表达明显低于正常组织，其低表达与肿瘤浸润深度及分期相关。Objective To investigate the expression of prostaglandin transporter （PGT） in colorectal cancer （CRC） tissues and its relationship with clinicopathological features. Methods The mRNA and protein levels of PGT were determined by real-time PCR , Western blot and immunohistochemical methods in cancer tissues and adjacent normal tissue from 80 patients with colorectal cancer and their relationship with clinicopathological features was analyzed. Results Compared with the adjacent normal tissue of colorectal cancer , the PGT mRNA relative expression （0.57 ±0.33 vs. 2.33 ±1.20） and the PGT protein expression in cancer tissues decreased significantly [PGT/GAPDH 0.45 ±0.16 vs. 0.78 ±0.23, integral A 718.7 ±359.4 vs. 10412.0 ±6423.3, average A 0.03 ±0.01 vs. 0.12 ±0.09, all P0.05）. Conclusion Expression levels of PGT mRNA and protein in colorectal cancer tissue are significantly down-regulation. PGT expression is associated with invasion depth and late stages.
