摘 要:用咪喹莫特诱发小鼠出现银屑病样改变,通过优化病理切片技术对皮损进行组织病理学研究.在Balb/c小鼠背部皮肤涂抹咪喹莫特7天,诱发银屑病样皮损,对其进行PASI评分,分析皮损程度;取皮损组织,制作石蜡病理切片,对浸蜡、脱水、染色等步骤进行温度、时长的优化,比较传统切片染色和优化方法对Baker组织病理学评分的影响.模型组小鼠涂抹咪喹莫特7天后出现红斑、鳞屑和皮肤增厚等银屑病样改变;镜下观察组织切片,发现其表皮增厚,角化不全,出现Munro小脓肿,棘层肥大等银屑病样病理变化.经本实验室常规方法处理的切片,组织不完整,结构不易分辨,影响组织病理学评分,从而影响对该动物疾病模型的鉴定;经改进处理后的切片,组织完整,结构清晰,染色均匀,能进行直观观察并准确评分,从而能准确鉴定银屑病疾病模型是否正确建立,为新药的临床前研究奠定基础.本实验成功制备银屑病样动物模型,通过切片和染色方法的改进获得较高质量的皮损组织石蜡切片,提高了疾病动物模型成功建立的鉴定标准,也为进一步的银屑病新药研究奠定了良好的基础.The psoriasis-like lesions were induced by imiquimod (IMQ)on BALB/c mice, which were observed and analyzed on the paraffin sections with the optimum paraffin section and dying methods. BALB/c mice received topical imiquimod on the shaved back for 7 consecutive days. On the 7th day, the severity of inflammation of the back skin was assessed based on the clinical Psoriasis Area and Severity Index(PASI). Then, the sample sections from the back skin were stained by HE for histology analysis with the optimum and traditional paraffin section and dying methods. Quantitative assessments of the pathological score, based on Baker' s method, were performed in five randomly chosen fields in each HE-stained section to assess the severity of psoriasis. The back skin of the mice displayed signs of erythema,scaling and thickening on the 7^th day after the IMQ application. At the same time, the epithelium hyperplasy, small Munro abscesses, dyskeratosis and acanthosis were observed on the sample sections and the Baker score was 5. 35 ± 0. 75, which was not observed on the sample sections treated with the traditional methods. With the optimum paraffin section and dying methods, the structural features of the skin were integrity, clear and the stain was uniform, which was beneficial for assessments of the pathological score on the skin. The high-quality paraffin sections were successfully prepared with the optimum paraffin section and dying methods. This method improved the identification of a diseased animal model and provided the basis for further research on the psoriasis treatment.
