摘  要:目的观察重组人内抑素(rh Endostatin)对兔耳增生性瘢痕成纤维细胞(HSFs)增殖的影响,探讨rh Endostatin抑制HSFs增殖的相关机制。方法制备兔耳增生性瘢痕(HS)模型,分离、培养并鉴定兔耳HSFs;应用四甲基偶氮唑盐(MTT)法和流式细胞仪分别检测rh Endostatin对HSFs增殖和周期的影响。结果分离培养的兔耳细胞经波形蛋白免疫细胞化学染色检测呈阳性。rh Endostatin对HSFs增殖的抑制效应与浓度和作用时间呈依赖关系,测得IC50值为100 mg/L。与正常对照组细胞相比,HSFs G1期细胞比例减少(P<0.01),S期和G2/M期细胞比例增加(P<0.01);与模型组细胞相比,rh Endostatin组细胞G1期比例增加(P<0.01),S期和G2/M期细胞比例减少(P<0.01)。结论rh Endostatin可抑制兔耳HSFs增殖,该作用可能与其引起HSFs细胞周期G1期阻滞相关。Objective To observe the effects of recombinant human endostatin ( rhEndostatin) on the proliferation of hypertrophic scar fibroblasts ( HSFs) derived from rabbit ears, and to explore the related mechanism of rhEn-dostatin inhibiting HSFs proliferation. Methods To establish the model of hypertrophic scar in rabbit ears, and i-solate, culture and identify HSFs of rabbit ears;MTT assay and flow cytometry were used to detect respectively the effects on proliferation and cell cycle of HSFs affected by rhEndostatin. Results The cells stained positive for vim-entin. rhEndostatin inhibited HSFs proliferation in a concentration- and time-dependent manner, and IC50 values measured for the 100 mg/L. The percentage of cells in G1 phase in HSFs decreased ( P〈0. 01 ) , and the rate of cells in S and G2/M phase increased (P〈0. 01), compared with that in normal control groups. The percentage of cells in G1 phase in HSFs treated with rhEndostatin increased ( P〈0. 01 ) , and the rate of cells in S and G2/M phase decreased ( P 〈0. 01 ) , compared with that in model groups. Conclusion RhEndostatin can inhibit the proliferation of hypertrophic scar fibroblasts derived from rabbit ears in vitro, which may be related with the G1 phase retardation in HSFs.
