摘 要:目的观察大鼠角膜、晶状体、视网膜中热休克蛋白A12B(heat shock protein A12B,HSPA12B)的表达。方法取SPF级SD大鼠9只,用水合氯醛处死后立即摘取眼球,采用Western blot和RT-PCR分别检测大鼠角膜、晶状体和视网膜组织中HSPA12B蛋白和mRNA的表达情况;免疫荧光化学法观察HSPA12B在角膜、晶状体和视网膜组织中的表达及定位。结果Western blot和RT-PCR检测结果显示:HSPA12B在大鼠角膜、晶状体、视网膜均有少量表达,其中角膜组织(0.280±0.034、0.374±0.031)中表达强度相对最低,晶状体(0.331±0.036、0.453±0.040)次之,视网膜组织(0.453±0.029、0.680±0.045)中表达强度最高。免疫荧光化学法结果表明,在角膜组织中HSPA12B主要表达于上皮层,其中基底细胞染色最强,而角膜基质细胞上仅有弱表达;在晶状体组织中HSPA12B主要表达于上皮细胞和皮质;在视网膜组织中HSPA12B主要表达于视网膜神经纤维层、神经节细胞层和内核层,并与NeuN标记的神经元及GS标记的Müller细胞存在共定位。结论HSPA12B在大鼠角膜、晶状体、视网膜组织中均有表达,且呈特异性表达。Objective To explore the expression of heat shock protein A12B( HSPA12B) in cornea, lens and retina tissue of rats. Methods The cornea, lens and retina tissue from 9 SPF SD rats were isolated. Western blot and RT-PCR assay were used to detect the protein and mRNA expressions of HSPA12 B in cornea, lens and retina tissue. Immunofluorescence staining was used to observe the location of HSPA12 B in cornea, lens and retina tissue. Results Western blot and RT-PCR analysis revealed that HSPA12 B were expressed in cornea, lens and retina tissue of rats.The expression of HSPA12 B was the weakest in cornea( 0. 280±0. 034,0. 374±0. 031),moderated in lens( 0. 331 ± 0. 036,0. 453 ± 0. 040),but it was strongest in retina( 0. 453 ± 0.029,0. 680±0. 045). Immunofluorescence staining showed that HSPA12 B mainly located at corneal epithelium with the strongest staining in the basal cells,but in contrast weak staining was seen in corneal stroma cells. HSPA12 B was strongly expressed in epithelial cells and shallow cortex layer of lens. In retina,HSPA12 B was extensively and richly expressed in retinal nerve fiber layer, ganglion cell layer and inner nuclear layer. The co-localization of HSPA12 B with the neuron marked by NeuN and the müller cell marked by GS in retinal tissue. Conclusion HSPA12 B is proved to express specificity in cornea, lens and retina tissue.
