摘  要:目的:探讨β-榄香烯诱导乳腺癌LCC-2细胞凋亡及逆转他莫昔芬耐药性的作用机制。方法:MTT法检测药物敏感性及耐药性逆转,流式细胞仪检测细胞周期和凋亡百分率,免疫组化SP法检测Bcl-2、pS2蛋白的表达。结果:β-榄香烯降低他莫昔芬对LCC-2细胞的IC50(P<0.01),逆转倍数为3.6倍;影响细胞周期时相,明显增强他莫昔芬对LCC-2细胞的凋亡诱导作用,凋亡率由(2.17±0.16)%上升至(9.13±0.34)%,(P<0.01);降低LCC2细胞Bcl-2表达,表达率由(56.52±4.85)%降至(28.02±6.32)%;pS2为阴性表达。结论:β-榄香烯(β-elemene)可部分逆转LCC-2细胞的耐药性,其逆转机制主要是抑制Bcl-2的表达,诱导耐药细胞凋亡。Objective:To study the effect of β-elemene on induction of apoptosis and reverses on tamoxifen (TAM)resistance in human breast cancer cell line LCC -2.Methods:The sensitivity of drug and the reversal of drug-resistance were examined by using MTT assay,the cell cycle and apoptosis were detected by flows cytometry, expression of Bcl-2 protein and pS2 protein were detected by SP method.Results:β-elemene could decrease the IC50 value of LCC-2 cell on tamoxifen,with a reversal of 3.6 fold,influence cell cycle,obviously strengthens.The effect of inducing apoptosis of LCC-2 cell.The apoptosis percentage was increased from (2.17 ±0.16)%to (9.13 ±0.34)%.The expression of Bcl-2 protein was reduced from (56.52 ±4.85)%to (28.02 ±6.32)%,expression of pS2 was negative.Conclusion:β-elemene could partially reverse tamoxifen resistance of LCC-2 cell line,its re-versal mechanism mainly related to suppress of the expression of Bcl-2,induces apoptosis of drug-resistance cell to reverses MDR.
