摘  要:目的建立昆明小鼠感染幽门螺杆菌(H.pylori)的动物模型。方法 32只SPF级昆明小鼠,随机分为模型组和对照组各16只。两组均给予200 mg/kg环磷酰胺腹腔注射,注射后第二天起模型组予以H.pylori SS1菌液灌胃,对照组给予等量脑心脊液灌胃,隔日1次,共5次,灌胃前禁食24 h,灌胃后禁食水2 h。末次灌胃2周、8周后两组分别处死8只小鼠,进行快速尿素酶试验(RUT)及HE、Warthin-Starry染色评估H.pylori定植率。结果模型组在末次灌胃菌液2周、8周后RUT阳性分别为5/8、8/8,对照组均为阴性;模型组HE染色下即可见到H.pylori定植,2周及8周定植成功数量均为8/8,对照组HE、Warthin-Starry染色下均为阴性。结论经免疫抑制预处理可成功建立H.pylori SS1感染昆明小鼠动物模型。Objective To establish the animal model by H. pylori infected mouse. Methods Thirty-twoSPF grade Kunming mice were randomly divided into model group and control group. Both groups were given 200 mg/kg CTX by intraperitoneal injection. The model group was inoculated with H. pylori SS1 while the control group with brain heart in- fusion once every other day and altogether five times. Both groups were fasting for 24 hours before inoculation and pro- hibited feeding and drinking for 2 hours. Eight mice in each group were sacrificed at 2 and 8 weeks respectively after the last inoculation. The H. pylori colonization was determined by rapid urease test (RUT) , HE and Warthin-Starry stai- ning. Results At the 2 weeks and 8 weeks after inoculation, the RUT showed positive results in moael group with 5/8 and 8/8 while the control group was negative. The H. pylori colonization was confirmed by HE staining in model group with both 8/8, and the control group was negative by HE or Warthin-Starry staining. Conclusion H. pylori SS1 infec- ted KM mice model can be successfully established after immunosuppression.
