摘　　要：目的探讨金属硫蛋白2A(MT2A)基因沉默对乳腺癌细胞呼吸链中细胞色素C氧化酶6B2亚基(Cpy6B2)表达的影响。方法乳腺癌野生细胞株MCF-7(以下简称MCF-7)分为空白对照组和MT2A干扰组,每组各12复孔。空白对照组不转染任何遗传物质,MT2A干扰组转染shRNA-MT2A载体,采用半定量PCR法检测两组MT2A mRNA的表达水平,MT2A干扰组成功获得MT2A基因沉默,采用实时定量PCR法分析两组Cpy6B2的表达水平。结果 MT2A干扰组中Cpy6B2的表达水平升高,与空白对照组相比,P<0.05。结论 MT2A基因可抑制人乳腺癌细胞Cpy6B2的表达。Objective To investigate the effect of metallothionein 2A（ MT2A） silencing on cytochrome C oxidase6B2 subunit（ Cpy6B2） expression of breast cancer cells. Methods The breast cancer wild cell line MCF-7（ hereinafter referred as the MCF-7） was divided into the blank control group and MT2 A interference group,and each group had 12 holes. The blank control group did not transfect any genetic materials,and the MT2 A interference group transfected the shRNA-MT2 A vector. Using semi-quantitative PCR to detect the MT2 A mRNA expression levels of the two groups. The MT2 A interference group successfully obtained the MT2 A gene silencing. The real-time quantitative PCR was used to analyze the expression of Cpy6B2 in the two groups. Results The results showed that the expression level of MCF-7 Cpy6B2 was increased under the MT2 A gene silencing. Conclusion MT2 A can inhibit the Cpy6B2 expression of breast cancer cells.
