摘 要:目的探讨多西环素对碱烧伤大鼠角膜新生血管的抑制作用及其机制。方法健康SD大鼠32只,随机分为对照组、多西环素组,每组16只。建立角膜碱烧伤模型后,多西环素组给予0.5%多西环素眼液点眼,对照组给予眼液溶媒点眼。分别于碱烧伤后3、7、14、21 d观察炎症指数、计算角膜新生血管面积;采用酶联免疫吸附试验检测角膜血管内皮生长因子(VEGF)表达。结果造模后多西环素组各时间点结膜充血、角膜水肿程度较对照组轻微;多西环素组3、7、14 d炎症指数均明显小于对照组(P<0.05);3、7、14、21 d角膜新生血管面积均小于对照组(P<0.05);两组大鼠角膜VEGF表达比较,差异无统计学意义(P>0.05)。结论多西环素可以减轻碱烧伤大鼠角膜的炎症反应及角膜新生血管形成,其机制可能与抑制炎症反应有关,而与VEGF表达无关联。Objective To investigate the inhibited effect of doxycycline on neovascularization ( NV) in a rat model of corneal alkali burn and its mechanism.Methods Thirty-two healthy SD rats were randomly divided into control group and doxycycline-treated group,with 16 rats in each group.After the corneal alkali burn model was established ,the doxycycline-treated group was given 0.5% doxycycline eye drop,while the control group was given the solution of eye drop without doxycycline .The inflammatory index was observed 3,7,14 and 21 days after injury ,respectively ,and the area of corneal NV was calculated .The cornea was detected for the expression of vascular endothelial growth factor(VEGF) by enzyme-linked immunosorbent assay(ELISA).Result s The doxycycline-treated group showed slighter conjunctival congestion and corneal edema at all time points compared with the control group .The inflammatory indexes 3,7,14 days after injury in the doxycycline-treated group were significantly lower than those in the control group (P&lt;0.05).The areas of corneal NV 3,7,14,21 days after injury in the doxycycline-reated group were smaller than those in the control group (P&lt;0.05).There was no significant difference in the expression of corneal VEGF between two groups (P&gt;0.05).Conclusion Doxycycline can relieve the corneal inflammatory reaction and corneal neovascularization in rats with corneal alkali burn ,which might be associated with the inhibition of inflammatory reaction ,but might not be associated with the expression of VEGF .
