丁基苯酞对蒙古沙土鼠全脑缺血再灌注损伤后海马组织p-ERK、Bcl-2及Bax蛋白表达的影响  被引量:1

摘  要:目的动态观察丁基苯酞(NBP)对蒙古沙土鼠全脑缺血再灌注损伤后海马组织形态学改变及p-ERK、Bcl-2及Bax蛋白表达变化。方法 42只蒙古沙土鼠随机分为三组:(1)正常对照组;(2)模型对照组(1 d、3 d、7 d);(3)NBP治疗组(1 d、3 d、7 d)。双侧颈总动脉夹闭10 min后再灌注,制备全脑缺血再灌注模型。Nissl染色观察海马CA1区锥体细胞形态及数量变化;Western blot蛋白印迹检测海马组织p-ERK、Bcl-2及Bax蛋白表达的动态变化。结果 Nissl染色光镜结果:(1)正常对照组:海马CA1区锥体细胞排列整齐,形态完整,未发现死亡的锥体细胞;(2)模型对照组:沙土鼠海马CA1区锥体细胞排列紊乱,胞体缩小、变形,核固缩,提示大多数锥体细胞变性及死亡,且随着全脑缺血再灌注时间的延长CA1区锥体细胞变性及死亡数目逐渐增加;(3)NBP治疗组:沙土鼠海马CA1区锥体细胞排列较整齐,形态较完整,细胞大而圆,尼氏体深染,与模型对照组比较锥体细胞数量明显增多,有显著性差异(P<0.01)。Western blot蛋白印迹检测结果:(1)p-ERK蛋白表达情况:模型对照组与正常对照组比较3 d p-ERK蛋白表达降低(P<0.05),1 d和7 d表达无改变,相对应时间点NBP治疗组p-ERK蛋白表达增加,且与模型对照组比较有显著性差异(P<0.05);(2)Bcl-2蛋白表达变化:模型对照组与正常对照组比较1 d和3 d Bcl-2蛋白表达降低(P<0.05),7 d表达无改变,相对应时间点NBP治疗组与模型对照组比较Bcl-2蛋白表达增加,有显著性差异(P<0.05);(3)Bax蛋白表达变化:模型对照组与正常对照组比较Bax蛋白在1 d、3 d、7 d表达均增加(P<0.05),且相对应时间点NBP治疗组与模型对照组比较Bax蛋白表达降低,有显著性差异(P<0.05)。结论丁基苯酞具有维持海马CA1区锥体细胞形态的完整性,减少锥体细胞变性及死亡的作用;丁基苯酞可能通过上调p-ERK、Bcl-2蛋白表达和下调Bax蛋白表达,抑制神经元凋亡,发挥保护作�Objective To detect the effect of dl-3n-butylphthalide(NBP) on the expression of p-ERK, Bcl-2, Bax and organization morphological changes in the hippocampus of Mongolian gerbil after global cerebral ischemia and reperfusion damage. Methods The forty-two Mongolian gerbils were randomly divided into normal control(CON) group, ischemia and reperfusion(I/R) group and NBP group.The bilateral common carotid arteries of Mongolian gerbil were clamped for 10 minutes and then released,prepared the whole cerebral ischemia reperfusion model. The morphological and quantitive changes on the pyramidal cells in the CA1 of hippocampus were observed by Nissl stain and the protein expression levels of p-ERK, Bcl-2, Bax in the hippocampus were detected by Western blot. Results The results of Nissl stain: in the CON group: the morphology of the pyramidal cells in the CA1 of hippocampus was intact and complete, no death pyramidal cells. The I/R group: the pyramidal cells in the CA1 of hippocampus in the I/R group were disorganized. The cell bodies were small and out of shape, karyopyknosis, most pyramidal cells degeneration and death, the CAl area pyramidal cell degeneration and death number increase as the longer of whole brain ischemia-reperfusion. In the NBP group: the pyramidal cells were well-arranged comparatively. The cells were round and big with the deeply-stained Nissl body and a small part of cytoplasm. The number of pyramidal cells were obviously increased compared with the I/R group, which was statistically significant(P〈0.01). The results of Western blot: the expression of p-ERK protein:compared with the CON group, the expression of p-ERK in the hippocampus in the I/R group was significantly down-regulated(P〈0.01), 1 d and 7 d expression without change, in the NBP group, the expression of p-ERK in the hippocampus was significantly up-regulated compared with the I/R group(P〈0.01); the expression of Bcl-2 protein: compared with the CON group, the expression of Bcl-2 in the hippoc
