摘  要:目的探讨1,25-二羟基维生素D3(1,25-(OH)2D3)对裸鼠宫颈癌移植瘤生长的抑制作用。方法制备BALC/c裸鼠皮下宫颈癌模型20只,随机分为4组:1,25-(OH)2D3组、单纯放疗组、1,25-(OH)2D3+放疗组及对照组,每组5只。观察各组裸鼠肿瘤的生长情况、裸鼠的一般情况,测量肿瘤直径并计算瘤体积。断颈处死裸鼠,天平称量肿瘤的重量。测定引流淋巴结中单个核细胞中CD4+、CD4+CD25+含量。结果 1,25-(OH)2D3组及单纯放疗组对肿瘤生长均有抑制作用,二者联合作用抑制更显著。1,25-(OH)2D3组、单纯放疗组及1,25-(OH)2D3+放疗组抑瘤率分别为34.8%、52.1%、54.1%。1,25-(OH)2D3干预后CD4+、CD4+CD25+含量与对照组及放疗组差异无统计学意义,而CD4+CD25+/CD4+在1,25-(OH)2D3组与对照组相比差异显著(P<0.05),1,25-(OH)2D3+放疗组与对照组相比差异显著(P<0.01)。结论 1,25-(OH)2D3+放疗对对裸鼠移植瘤宫颈癌生长有显著免疫抑制作用,为宫颈癌的综合治疗提供了实验依据。Objective To study the inhibition of 1,25-dihydroxyvitamin D3( 1,25-( OH)2D3)against the transplanted cervical cancer on nude mice. Methods Twenty BALC / c athymic mice uterine cervix cancer analogues were divided randomly into 4 groups: radiation 10 Gy group,1,25-( OH)2D3group,1,25-( OH)2D3+ radiation 10 Gy group and control group,5mice in each group. The tumorous growth,general state of health and life span were observed in all groups,tumor diameter were measured and tumor volume were calculated. The mice were sacrificed,the tumor weight were determined. Mononuclear cells( MNCs) in draining lymph nodes were collected,then numbers of CD4+CD25+cells were determinated by flow cytometry.Results Radiation 10 Gy group and 1,25-( OH)2D3group and 1,25-( OH)2D3+ radiation 10 Gy group had depressant effect on tumor growth. The volume and weight of transplanted tumor in experimental group were significant inhibited compared with control group( P 0. 05). The inhibition ration of tumor of 1,25-( OH)2D3group,radiation 10 Gy group and 1,25-( OH)2D3+ radiation 10 Gy group were 34. 8%,52. 1% and 54. 1%. There was no significant difference in number of CD4+CD25+T cells of MNCs in draining lymph nodes between the treated group and the control group; The CD4+CD25+/ CD4+in the 1,25-( OH)2D3revented group were significantly higher than those in the control group( P 0. 05). Conclusion Administration of 1,25-( OH)2D3+ radiation 10 Gy can decrease uterine cervix cancer transplantation tumor,It may afford experimental basis for the uterine cervix cancer combined therapy.
