摘　　要：目的:探讨趋化因子CXCL4在结直肠癌组织中的临床意义。方法:160例结直肠癌患者根据国际抗癌联盟(UICC)的TNM分期分为Ⅰ、Ⅱ、Ⅲ、Ⅳ期,采用RT-PCR方法定量检测CXCL4 m RNA的表达,应用免疫组织化学法和Western blot检测结直肠癌组织及癌旁正常组织CXCL4蛋白表达水平,探讨CXCL4的表达与结直肠病理因素之间的关系。结果:CXCL4 mRNA在结直肠癌组织中的平均相对表达量低于癌旁正常组织,差异具有统计学意义(P<0.05)。CXCL4蛋白在结直肠癌组织中的阳性表达率为86.2%(138/160)。在结直肠组织中,CXCL4的表达与肿瘤大小、分化程度、淋巴结转移、TNM分期相关(P<0.05)。结论:趋化因子CXCL4与结直肠癌的一些临床病理因素密切相关,可能成为评估结直肠癌预后的生物学指标之一。Objective： To detect the expression of CXCL4 in colorectal cancer and explore the significance. Methods：160 colorectal cancer Patients were divided into four groups（I, II, III, IV, re-spectively）. The m RNA expression of CXCL4 in these tissue specimens was measured by reverse transcription PCR. The expression of CXCL4 was detected by immunohistochemistry and Western-Blot in 160 primary lesions of colorectal cancer and normal colorectal tissues adjacent to cancer lesion, analyzing the correlation between clinicopathological factors of patients and expression of CXCL4 protein. Results： The expression level of CXCL4 m RNA in colorectal cancer tissues was significantly lower than that in matched tumor-adjacent colorectal tissues（P0.05）.The positive rate of CXCL4 protein expression in colorectal cancer tissues was 86.2%. The expression of CXCL4 protein was closely related to size, histological differentitation, lymph node metastasis, TNM stage（all P0.05）. Conclusion： The expression of CXCL4 was significantly correlation with malignant behaviors of CXCL4,thus CXCL4 might be an important biological marker in the oncogenesis, progression, biological behavior and prognosis of colorectal cancer.
