摘 要:目的:以ERα(+)子宫内膜癌细胞株Ishikawa为研究对象,建立子宫内膜癌细胞的裸鼠皮下移植瘤模型,进一步研究在子宫内膜癌细胞移植瘤中ERK、p-ERK、Akt、p-Akt蛋白的表达及其意义。方法:利用转染前后的Ishikawa细胞建立子宫内膜癌细胞裸鼠皮下移植瘤模型,采用免疫组织化学链霉亲和素-生物素过氧化物酶复合物(SABC法)检测裸鼠移植瘤中ERRγ蛋白的表达情况。Western blot方法检测移植瘤组织中Akt、p-Akt、ERK、p-ERK蛋白的表达情况。结果:免疫组化检测Ishikawa细胞移植瘤组与no-silence Ishikawa细胞移植瘤组(空载体组)ERRγ蛋白表达阳性率无显著性差异(P>0.05),Ishikawa细胞移植瘤组与ERRγ-shRNA Ishikawa细胞移植瘤组阳性率具有显著性差异(P<0.05)。Western blot方法检测3组组织的活化情况:ERRγ-shRNA Ishikawa细胞移殖瘤组中Akt、p-Akt、ERK、p-ERK表达量均低于Ishikawa细胞移殖瘤及no-silence Ishikawa细胞移殖瘤组(P<0.05)。结论:沉默ERRγ基因的瘤组织的ERK及Akt的表达呈现低表达,从而可以推测:通过ERRγ的表达来调节ERK及Akt信号通路的表达,进一步抑制子宫内膜癌Ishikawa细胞增殖,促进其凋亡。Objective:To use ERα ( + ) Ishikawa endometrial cancer cell line as the research object,to establish endometrial cancer nude mice, and research ERRγ endometrial cancer ERK, p - ERK, Akt, p - Akt protein and its significance. Methods:Immunohistochemical strept avidin -biotin -biotin peroxidase complex (SABC) was used to detect growing tumor in nude mice ( Ishikawa tissue, expression ERRγ- shRNA Ishikawa ceils and tissues, and trans- ferred to the empty vector no - silence Ishikawa tissue) in ERRγ protein expression. Western blot method was used to detect Akt, p - Akt, ERK, p - ERK protein activation. Results: In Ishikawa ceils transplanted tumor group of no - si- lence the Ishikawa cell transplant tumor group positive rate was not significantly significant different ( P 〉 0.05 ). Ish- ikawa cells transplanted tumor group ERRγ - shRNA Ishikawa cells transplanted tumor group positive rate had a sig nificant difference ( P 〈 0.05 ). The ERRγ - shRNA Ishikawa cell transplants tumor group, Akt, p - Akt, ERK, p - ERK expression were lower than Ishikawa cell transplant tumor and no - silence Ishikawa cell transplant tumor group ( P 〈 0.05 ). Conclusion: After ERRγ gene silencing tumor tissue expression of ERK and Akt showed low expression. ERRγ expression can influence the expression of ERK and Akt, ERR5, involved in the ERK and Akt signaling path- ways, Ishikawa endometrial cancer inhibit cell proliferation, induce apoptosis, ERRγ in endometrial cancer occurrence and development play an important role.
