摘 要:目的:研究FoxM1在胶质瘤细胞对卡氮芥化疗耐药中的作用,为胶质瘤化疗提供新的靶点和依据。方法:建立对BCNU耐药的胶质瘤细胞株,采取Western blot法检测FoxM1蛋白表达水平的变化;通过RNA干扰技术下调FoxM1表达水平,CCK-8法检测胶质瘤细胞对卡氮芥的敏感性;检测DNA损伤修复基因XRCC1蛋白表达水平。结果:卡氮芥耐药的U251胶质瘤细胞中FoxM1与XRCC1的蛋白表达水平明显升高;下调FoxM1表达水平后,恢复了耐药细胞株对卡氮芥的敏感性;下调FoxM1蛋白表达水平后,XRCC1蛋白表达水平随之下降,与胶质瘤细胞对卡氮芥的敏感程度相关。结论:FoxM1在胶质瘤细胞对卡氮芥耐药过程中发挥重要作用;在这一过程中,DNA损伤修复机制参与了FoxM1介导的胶质瘤细胞对卡氮芥的耐药过程。Objective:To study the role of FoxM1 on BCNU resistance in glioma cells, provides a new target and basis for glioma chemotherapy. Methods: BCNU - resistant glioma cell line was established, the protein expression level of FoxM1 was detected by Western blot. The FoxM1 expression level was reduced by RNA interference, the sen- sitivity of glioma cells to BCNU was detected by CCK - 8 assay. The protein expression level of XRCC1 was detected by Western blot. Results:The expression levels of FoxM1 and XRCC1 were significantly increased in BCNU -resist- ant U251 cells. The sensitivity to BCNU in BCNU - resistant U251 cells was restored by reduce the expression of FoxM1. The protein expression level of XRCC1 was reduced by shRNA - FoxM1 ,associated with the sensitivity of gli- oma cells to BCNU. Conclusion:FoxM1 play an important role in the process of BCNU resistance in glioma cells, DNA damage repair mechanisms is involved in the BCNU resistance mediated by FoxM1 in glioma cells.
