摘 要:目的:研究蓝莓对酒精性脂肪肝(AFLD)小鼠肝组织中血红素氧化酶-1(HO-1)表达水平及抗氧化能力的影响,探讨蓝莓对AFLD的保护作用及可能机制。方法:40只C57/6J小鼠随机均分为对照组、蓝莓组、蓝莓+HO-1抑制剂(锌原卟啉,Zn PPIX)组及模型组,NIAAA法建立AFLD动物模型,油红O染色法检测模型建立情况及各组小鼠肝组织病理学变化;取眼眶血检测各组小鼠血清谷丙转氨酶(ALT),谷草转氨酶(AST),甘油三脂(TG),胆固醇(TC)含量;酶联免疫吸附法(ELISA)测定各组小鼠肝组织中超氧化物歧化酶(SOD)活性及谷胱甘肽(GSH)、丙二醛(MDA)的含量;Western blot法检测各组小鼠肝组织中HO-1蛋白的表达。结果:蓝莓组肝组织脂肪沉积较模型组及蓝莓+Zn PPIX组明显减少,血清中ALT、AST、TG及TC含量降低(P<0.05),肝组织中SOD活性和GSH含量升高(P<0.05),MDA含量降低(P<0.05);蓝莓组HO-1蛋白表达量高于蓝莓+ZnPPIX组及模型组。结论:蓝莓可减轻AFLD,其机制可能与上调小鼠肝脏HO-1的表达,增强机体抗氧化能力有关。Objective: To investigate the influence of blueberry on heme oxygenase-1( HO-1) and antioxidant ability of mice with alcoholic fatty liver disease( AFLD),and to explore the protective effects of blueberry on AFLD and possible mechanism. Methods: Forty C57 /6J mice were randomly divided into control group,blueberry group,blueberry + Zn PPIX group and model group. AFLD model was established by using NIAAA method,pathological changes in the hepatic tissues were detected by oil red O staining,the orbital blood was taken to detect the levels of alanine aminotransferase( ALT),aspartate aminotransferase( AST),triglyceride( TG) and total cholesterol( TC). The superoxide dismutase( SOD) activity and glutathione( GSH) levels,malondialdehyde( MDA) contents inliver tissue were quantified by ELISA. The expression of HO-1 in liver tissue was detected by Western blot. Results: Compared with model group and blueberry + Zn PPIX group,lipid accumulation in liver tissue of mice in blueberry group decreased,serum ALT,AST,TG and TC levels decreased( P〈0. 05). The hepatic MDA content also decreased( P〈0. 05),while GSH contents,SOD activity and the HO-1levels increased( P〈0. 05). Conclusion: Blueberry can alleviate AFLD,the mechanism may relate to raise the expression of HO-1 in mice,increase the antioxidant capacity.
