摘 要:目的:探讨间变性淋巴瘤激酶(ALK)基因在卵巢癌组织中的表达。方法:选取61例卵巢上皮性癌组织和10例良性卵巢上皮性肿瘤组织、10例正常卵巢组织,应用免疫组织化学方法检测ALK蛋白,ALK阳性者用FISH进行进一步的确认。结果:卵巢良性上皮性肿瘤组织及正常卵巢组织不表达ALK蛋白,ALK基因在卵巢癌组织中的表达阳性率为1.64%。在61例卵巢上皮性癌中,只发现1例年龄超过50岁、临床Ⅰ期和无腹膜转移的ALK蛋白阳性,经FISH确认为EML4-ALK基因阳性。结论:ALK基因可能在卵巢癌的发生发展过程中发挥着协同作用,提示EML4-ALK有可能成为卵巢癌的一种亚型,作为新的靶向治疗指标。Objective: To explore the expression of anaplastic lymphoma kinase( ALK) gene in epithelial ovarian cancer. Methods: Immunohistochemical method was used to detect ALK proteins expressions in 61 cases with epithelial ovarian cancer,10 cases with benign epithelial ovarian tumor,and 10 cases with normal ovary; the cases with positive ALK were further determined by FISH. Results: ALK protein did not express in benign epithelial ovarian tumor and normal ovary,the positive expression rate of ALK gene in epithelial ovarian cancer was 1. 64%. Among 61 cases with epithelial ovarian cancer,only one case was more than 50 years old; ALK protein expressed positively in the cases of clinical stage Ⅰ and without peritoneal metastasis,then they were determined as positive EML4- ALK gene definitely by FISH. Conclusion: ALK gene may have synergistic effect in oncogenesis and development of ovarian cancer,which indicates that EML4- ALK may become to be a subtype of ovarian cancer,which can be used as a new index for targeted therapy.
