摘 要:为了研究细粒棘球绦虫nanos基因的序列及结构并为疫苗及新的药物研制提供靶标,试验采用注释并克隆细粒棘球绦虫nanos基因的方法,并对其序列结构进行分析;利用在线生物信息学软件分析细粒棘球绦虫nanos基因的序列结构及与其他虫属的进化关系。结果表明:细粒棘球绦虫nanos基因ORF为687 bp,编码228个氨基酸,理论分子质量为58 090.6 u,等电位点为5.08;细粒棘球绦虫NANOS蛋白由3个α螺旋和4个β片层组成;发现共有1个丝氨酸位点,1个苏氨酸位点,2个酪氨酸位点,可能的磷酸化位点是7,8,13,18,19,23位。说明该基因可能在细粒棘球绦虫生殖细胞发育过程中发挥着重要作用。To study the sequence and structure of the nanos gene from Echinococcus granulosus( E. granulosus),and provide the targets for the development of vaccines and new drugs. The nanos gene from E. granulosus was noted and cloned,and its sequence structure was analyzed.The sequence structure of the nanos gene from E. granulosus and the evolutionary relationshipsnanos with other genus were analyzed using online bioinformatics software. The results showed that the ORF of the nanos gene from E. granulosus was 687 bp,encoding a protein of 228 amino acids with a molecular weight of 58 090. 6 u,and the isoelectric point was 5. 08. The NANOS protein from E. granulosus was composed of threeα helixes and four β- sheet layers. It was found that there were one serine site,one threonine site,and two tyrosine sites,and the potential phosphorylation sites were 7,8,13,18,19,and 23 positions. The results indicate that the nanos gene may play an important role in the process of germ cell development in Echinococcus granulosus.
