血必净注射液对脓毒症大鼠心功能的影响

摘　　要：目的：观察血必净注射液对盲肠结扎穿孔术（ CLP）大鼠心功能的改善作用。方法清洁级雄性Wistar大鼠随机分为正常组、假手术组、CLP组和血必净组，后三组按留取标本时间分为术后12、24、48和72 h四个亚组，每组6只大鼠。各组于相应时间点通过超声心动图测定大鼠左心室射血分数（ LVEF），经腹主动脉取血，采用酶联免疫吸附试验（ ELISA）检测血浆心房脑钠肽（ BNP）、肿瘤坏死因子－α（ TNF－α），留取心肌组织观察病理改变。结果 CLP组大鼠较对照组及假手术组LVEF降低（P＜0．05），BNP明显升高（P＜0．01），病理评分升高（P＜0．05）， TNF－α明显升高（P＜0．01）；血必净组大鼠LVEF升高（P＜0．05），BNP、病理评分及TNF－α明显降低（ P＜0．05或P＜0．01）。结论血必净注射液可降低脓毒症大鼠TNF－α和BNP水平，改善心肌病理评分及心功能障碍。Objective To investigate the effects of Xuebijing injection on cardiac function in cecal ligation puncture （CLP ） rats.Methods Clean healthy male Wistar rats were randomly divided into normal control group, sham operation group, CLP group and Xuebijing （XBJ） treated group.The three latter groups were divided into four groups of 12, 24, 48 and 72-hour subgroups with 6 rats in each one.Left ventricular ejection fraction （ LVEF ） of the rats were detected by echocardiography at various time points. The blood samples of each group were collected at various time points from abdominaly aorta.Serum levels of brain natriuretic peptide （ BNP ） and tumor necrosis factor -α（ TNF-α） were detected by euzymelinked immunosorbent assay （ ELISA） .The sample of the cardiac muscle were harvested aseptically.The histopathologic changes of myocardial tissue were observed. Results Compared with normal control and sham operation group LVEF decreased （P〈0.05）; BNP markedly increased （P〈0.01）, the myocardial histopathologic score was increased （P〈0.05） and TNF-αmarkedly increased （ P〈0.01） in CLP group,and those levels were markedly improved in the Xuebijing-treated group （P〈0.05或P〈0.01）.Conclusion Xuebijing injection could decrease the CLP rats TNF -αand BNP, and obviously ameliorate the myocardial histopathologic score and the cardiac dysfunction.
