摘 要:目的通过构建自噬相关基因ATG5沉默的人鼻咽癌低分化鳞癌细胞(CNE-2细胞)裸鼠移植瘤模型,研究自噬与鼻咽癌放射敏感性的关系。方法选取24只裸鼠,随机分入对照组、NC组和ATG5组,每组8只。对照组裸鼠接种CNE-2细胞,NC组裸鼠接种转染阴性对照载体的CNE-2细胞,ATG5组裸鼠接种稳定沉默ATG5基因的CNE-2细胞。每组各取4只裸鼠给予10Gy射线照射,观察各组裸鼠移植瘤的生长情况,并应用Western印迹法检测自噬相关蛋白ATG5和LC3Ⅱ的表达。结果自噬相关基因ATG5沉默的人鼻咽癌CNE-2细胞裸鼠移植瘤模型构建成功,成瘤率为100%。ATG5组的LC3Ⅱ、ATG5蛋白相对表达量显著低于对照组和NC组(P值均<0.05),ATG5+10Gy组也均显著低于对照+10Gy组和NC+10Gy组(P值均<0.05)。结论自噬相关基因ATG5沉默可以增加鼻咽癌CNE-2细胞裸鼠移植瘤的放射敏感性,推断自噬与鼻咽癌放射敏感性呈负相关。Objective To construct mouse models with transplanted nasopharyngeal carcinoma cell (CNE-2) containing silent autophagy-related gene 5 (ATG5), and to investigate the effect of autophagy on radiosensitivity of nasopharyngeal carcinoma. Methods A total of 24 nude mice were randomly divided into 3 groups; control group, NC group and ATG5 group. There were 8 mice in each group. CNE-2 cells were transplanted in the control group. CNE-2 cells transfected with empty vectors were transplanted in the NC group. CNE-2 cells containing silent ATG5 gene were transplanted in the ATG5 group. Four mice of each group were exposed to 10 Gy X-ray. The tumor growth was observed in each group. The expression of LC3 g and ATG5 protein was measured by Western blot. Results The xenograft tumor model was established successfully. The expression of ATG5 and LC3-Ⅱ protein in the ATG5 group was significantly lower than that in the control group and NC group (all P〈0.05). After irradiation by 10 Gy X-ray, the expression of ATG5 and LC3-Ⅱ protein in the ATG5 group was also significantly lower than that in the other two groups (all P〈0.05). Conclusion ATG5 gene silencing can enhance the radiosensitivity of tumors in nude mice implanted with CNE-2 cells. Autophagy may be negatively correlated to the radiosensitivity of nasopharyngeal carcinoma.
