舌鳞癌酸性微环境下雷公藤多甙对单核-巨噬细胞分泌血管内皮生长因子的影响

摘  要:目的检测雷公藤多甙(tripterygium wilfordii polyglycosidium,TWP)在模拟舌鳞癌局部酸性微环境下对单核-巨噬细胞分泌血管内皮生长因子(vascular endothelial growth factor,VEGF)的影响。方法在p H值6.6、6.8酸性微环境中,用对数生长期的Tca8113细胞培养上清液与人单核-巨噬细胞系THP-1细胞混合培养,经不同浓度的TWP干预及干预不同时间,以酶联免疫吸附测定法检测其对THP-1细胞分泌VEGF的影响。结果 THP-1细胞与Tca8113细胞上清液混合培养4 h,THP-1细胞分泌VEGF受到抑制,分泌抑制率随TWP浓度增加而增大,与浓度成正相关(p H值6.6时,r=0.983,P<0.001;p H值6.8时,r=0.971,P<0.001)。p H值为6.6,TWP浓度为5×10-1μg/m L时,分泌VEGF抑制率随时间延长而增大,与时间成正相关(r=0.975,P<0.05)。结论体外模拟的舌鳞癌酸性微环境中,TWP抑制单核-巨噬细胞分泌VEGF,抑制率分别与药物浓度及作用时间正相关。Objective To investigate the effects of triptergium wilfordii polyglycosidium on the secretion of vascular endothelial growth factor from mononuclear-macrophages in acid oral squamous-cell carcinoma microenvironment. Methods ELISA assay was used to evaluate the effects on the secretory of VEGF in THP-1 monocytes, co-cultured with su- peroate of Tca8113 cells' in exponential phase of growth, in vitro acid microenvironments (pH6.6,6.8) , treated by dif- ferent concentration of TWP. Results ELISA assay showed that the secretory of VEGF of THP-1 monocytes had been significantlydecreased, after treated with TWP, co-cultured with the supernate of Tea8113 cells' in exponential phase of growth, in the identical vitro acid micro-environments ( pH6.6 or 6.8 ) ( P 〈 0.05 ). The ischesis ratio had been significantly increased with the raising of the densities of TWP, and presented significantly positively related to the concentration (P 〈 0.05 ). The ischesis ratio had also been significantly increased with time (P 〈 0.05 ), at pH6.6, interfered by TWP of 5 × 10^ -1 μg/mL, and presented significantly positively related to time ( P 〈 0.05 ). Conclusion The secretory of VEGF of the mononuclear-macrophages interfered by TWP had been significantly decreased , which were cultured in vitro acid tongue squamous-cell carcinoma microenvironment, and the ischesis ratio presented significantly positively related to the concentration and time.
