摘 要:目的探讨盐酸多奈哌齐对血管性痴呆(VD)小鼠学习记忆能力及对海马Akt、p-Akt表达的调节作用。方法采用双侧颈总动脉反复缺血-再灌注法制备小鼠VD模型。90只雄性昆明小鼠随机分为3组,假手术组(30只)、模型组(30只)及盐酸多奈哌齐组(30只)。在术后第29天、第30天,经跳台试验和水迷宫试验进行行为学测试,用免疫组化和western blot方法检测各组小鼠海马Akt、p-Akt的表达变化。结果小鼠的学习和记忆能力VD模型组明显低于假手术组,而盐酸多奈哌齐治疗组明显高于VD模型组小鼠(<0.05);免疫组化和Western blot结果发现,小鼠海马CAl区Akt、p-Akt蛋白相对表达水平VD模型组明显低于假手术组,而盐酸多奈哌齐治疗组显著高于VD模型组(<0.05)。结论盐酸多奈哌齐改善血管性痴呆(VD)小鼠学习和记忆功能可能与上调VD小鼠Akt、p-Akt的表达相关。Objective To study the effect of donepezil on learning and memory and the expression of Akt and p-Akt protein in hippocampus of mice with vascular dementia(VD). Methods 90 Kunming mice were randomly divided into sham group(30), model group(30)and treatment group(treated with donepezil, 30). The models of VD mice were established by three times ischemia-reperfusion of bilateral common carotid arteries. The behavioral scores were investigated by step-down test and water maze test at days 29, 30 respectively. The expressions of Akt and p-Akt were examined in hippocampus of mice by immunohistochemistry and Western blot methods. Results The learning and memory abilities were obviously decreased in model group than in sham group, but increased in donepezil treatment than in VD mice(P〈 0.05). The expression levels of Akt and p-Akt in hippocampus were decreased in model group than in sham group, but higher in donepezil treatment than in VD mice(P〈 0.05). Conclusion The improvement of learning and memory abilities by donepezil might be related to the upregulation of Akt and p-Akt expression in VD mice.
