miR-16及PTEN蛋白在子宫内膜癌中的表达及其意义

摘　　要：目的探讨miR-16mRNA及PTEN蛋白和mRNA在子宫内膜癌(EC)组织中的表达及意义。方法实时定量PCR方法检测子宫内膜癌组织miR-16mRNA及PTEN mRNA的表达水平,Western blot方法检测子宫内膜癌组织PTEN蛋白表达;统计分析子宫内膜癌组织中miR-16与PTEN蛋白表达的相关性。结果与癌旁正常组织相比,miR-16mRNA在子宫内膜癌组织中的表达水平明显升高(<0.01),PTEN蛋白和mRNA表达水平明显降低(<0.01),二者呈显著地负相关关系。结论 miR-16与PTEN在子宫内膜癌组织表达呈显著的负相关。Objective To study the expressions of miR-16 mRNA and PTEN protein and mRNA in endometrioid carcinoma（EC）tissues. Methods The expression of mi R-16 mRNA and PTEN mRNA in EC tissues was detected by real-time quantitative PCR, the expression of PTEN protein was detected by Western blot method. The correlation analysis was performed for investigating the relationship between mi R-16 and PTEN expressions. Results Compared with normal tissues, the expression level of mi R-16 mRNA was significantly increased in EC tissues（ P〈0.01）, but the expression level of PTEN protein and PTEN mRNA was significantly downregulated in EC tissues than in normal tissues（ P〈0.05）. The expression of PTEN mRNA in EC tissues was negatively correlated with the expression of mi R-16 mRNA. Conclusion The expression of mi R-16 in endometrial carcinoma was upregulated, but downregulated for PTEN m RNA.
