TGF-β1对rhBMP-2促进大鼠骨髓间充质干细胞增殖分化的影响

摘　　要：目的观察转化生长因子β1(TGF-β1)对重组骨形态蛋白2(rhBMP-2)促进大鼠骨髓间充质干细胞增殖分化的影响。方法分离培养骨髓间充质干细胞,然后加入不同浓度的rhBMP-2,用MTT法检测细胞增殖情况,通过ELISA法检测细胞碱性磷酸酶(ALP)活性评估其骨向分化能力。同时加入TGF-β1观察其对rhBMP-2协同促进能力。结果不同浓度rhBMP-2对骨髓间充质干细胞(MSCs)进行处理后检测出的吸光度值(D值)及ALP活性检测值均较阴性对照组明显升高(P<0.05),随着浓度增加,其促进作用有所提高。而TGF-β1+rhBMP-2组检测到的吸光度值及ALP活性升高更明显。结论 TGF-β1能够有效促进rhBMP-2对MSCs的增殖和成骨分化作用。[ ABSTRACT] Objective The research was to investigate the effect of transforming growth factor-β1 （ TGF-β1 ） on the growth and proliferation of the Mesenchymal stem cells after recombinant human bone morphogenetic protein -2 （rhBMP-2） stimulating.Methods MSCs were isolated from bone marrow of male Sprague-Dawley（SD） rats and ad-hered to the culture plate in vitro.MSCs were divided into 3 groups as follows：（Group 1） control,（Group 2）rhBMP-2 in different concentrations,and（Group 3） TGF-β1＋rhBMP-2 in different concentration.MTT method was used to confirm the most effective concentration of rhBMP-2 on the proliferation of MSCs .ALP activity was evaluated by ELISA test .Re-sults The number of MSCs and the ALP activity increased with rhBMP-2 in a concentration-dependent manner .A sig-nificantly increased number of MSCs was found at TGF-β1＋rhBMP-2 group（P〈0.05）.Conclusion The findings sug-gest that TGF-β1＋rhBMP-2 or rhBMP-2 alone promote the osteoblastic differentiation of MSCs .
