卡介苗多糖核酸对口腔扁平苔藓病损区IL-12p40表达的影响

摘　　要：目的探讨卡介苗多糖核酸(BCG-PSN)对口腔扁平苔藓(OLP)病损区培养角质细胞分泌细胞因子IL-12p40的影响和意义。方法取经病理诊断为口腔扁平苔藓病损组织做细胞培养:分为OLP组、OLP+BCG-PSN组,每组6例,在角质形成细胞长满时加入BCG-PSN,用同等体积生理盐水作为阴性对照,在培养6h、12h、24h、48h分别收集培养细胞上清液,ELISA法检测IL-12p40浓度变化情况。结果 1整个细胞生长期间为单一的角质形成细胞,细胞为椭圆形或多角形,呈现典型的铺路石状,符合上皮角质形成细胞生长形态特征。2在不同的培养时段内,BCG-PSN共同培养刺激角质形成细胞产生IL-12p40的减少量与对照组相比均有显著差异(6h F=7.87 P=0.0186,P<0.05;12h F=10.31 P=0.0093 P<0.05;24h F=16.57 P=0.0022 P<0.05;48h F=17.3 P=0.002 P<0.05)。结论 BCGPSN可能通过对OLP角质形成细胞分泌IL-12p40有直接的抑制效果起到减轻T细胞引起的直接损伤、降低局部自身免疫反应的作用。Objective To investigate the effect and significance of the polysaccharide nucleic acid fraction of bacillus calmette guerin to keratinocyte cultured in vitro secretion of IL-12p40 on patients with oral lichen planus（OLP）.Methods The OLP epithelial cells by pathological diagnosis were divided into OLP group and OLP＋ BCG-PSN group, 6cases of BCG-PSN were added almost the cell covered, control group were added the same volume of saline. In cultured6 h, 12 h, 24 h, 48 h, the cell culture supernatant were collected to detect IL-12p40 concentration by ELISA method.Results 1During the entire cell growth for single keratinocytes, cells were oval or polygonal, showed cobblestone like typical epithelial keratinocyte morphological characteristics. 2 There was significant difference that concentration of IL-12p40 co-cultured with BCG-PSN stimulation of keratinocytes produced decreased compared with control group in different culture time（6h F=7.87 P=0.0186, P〈0.05; 12 h F=10.31 P=0.0093 P〈0.01; 24 h F=16.57 P=0.0022P〈0.01; 48 h F=17.3 P=0.002 P〈 0.01）. Conclusion BCG-PSN have direct inhibiting effect to T cells, reduce the local autoimmune reaction through inhibiting secretion of IL-12p40 in cultured keratinocytes of OLP.
