当归注射液对椎板切除术后兔模型硬膜外瘢痕中PDGF表达的影响

摘　　要：目的:探讨当归注射液对椎板切除术后硬膜外粘连及瘢痕形成兔动物模型的预防作用和干预机制,为临床预防和治疗腰椎手术失败综合症(FBSS)提供理论依据。方法:选取成年健康新西兰大白兔45只分为3组,每组15只,分别为模型组(生理盐水组)、对照组(玻璃酸钠组)、实验组(当归注射液组)。于术后2周、4周、6周分3批处死白兔,从组织学观察硬膜外粘连和测定瘢痕中血小板衍化因子(PDGF)含量。结果:第2周,3组比较,各组间比较差异无统计学意义(P>0.05)。第4周,模型组与对照组比较,P<0.05,模型组与实验组比较,P<0.05,差异均有统计学意义。而对照组与实验组比较差异无统计学意义(P>0.05)。第6周,模型组与对照组比较,P<0.05,模型组与实验组比较,P<0.05,差异均有统计学意义,而对照组与实验组比较差异无统计学意义(P>0.05)。说明当归注射液对预防硬膜外粘连有一定的效果,但与玻璃酸钠相比却无明显优势。结论:当归注射液可以减少FBSS兔局部成纤维细胞计数及PDGF含量的增加。Objective：To investigate the preventive effects and interventional mechanisms of Danggui injection on rabbits＇epidural adhesion and scar formation after laminectomy,and to provide theoretical basis for clinical prevention and treatment of failed back surgery syndrome（FBSS）.Methods：Adult healthy New Zealand white rabbits were divided into three groups,each group of 15 and listed as model group A,control group B and trial group C.Rabbits were executed 2weeks,4weeks and 6weeks after operation respectively.Epidural adhesion was observed by hyphology and platelet derived growth factor（PDGF）expression was measured in scar.Results：There were no significant differences among three groups 2weeks after the operation（P〉0.05）.There were significant differences between group A and group B,also group A and group C 4weeks after operation（P〈0.05）,but not between group B and group C（P〉0.05）.There were significant differences between group A and group B,also group A and group C 6weeks after the operation（P〈0.05）,and also between group B and group C（P〉0.05）.Above all,Danggui injection has effect on prevention of epidural adhesion,but it has no obvious advantage compared with the sodium hyaluronate.Conclusion：Danggui injection decreases the number of fibroblasts and the rising PDGF expression of FBSS rabbits.
