摘　　要：目的探讨E-钙黏附蛋白(epithelia-cadherin,E-cadherin)和E盒结合锌指蛋白2(E-box binding zinc finger protein 2,ZEB2)在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的表达及预后意义。方法采用免疫组织化学SP法检测E-cadherin蛋白和ZEB2蛋白在144例NSCLC患者肺癌组织(NSCLC组)及52例NSCLC患者癌旁>5cm的正常肺组织(对照组)中的表达情况,应用Cox回归模型分析预后影响因素。结果NSCLC组ZEB2蛋白表达率(78.5%)高于对照组(11.5%),而E-cadherin蛋白表达率(34.0%)低于对照组(100.0%),差异均有统计学意义(P<0.05);NSCLC组ZEB2、E-cadherin蛋白阳性表达率在肿瘤分化程度、TNM分期和淋巴结转移上差异有统计学意义(P<0.05);NSCLC组ZEB2与E-cadherin蛋白表达呈负相关性(r=-0.444,P=0.000);多因素Cox回归分析显示ZEB2、E-cadherin的表达水平可作为NSCLC预后的独立因素。结论 ZEB2在NSCLC中表达上调,致使E-cadherin蛋白表达下调,从而使肿瘤浸润和转移能力增强,最终导致预后不良。Objective To explore the expressions of epithelia-cadherin（E-cadherin）and E-box binding zinc finger protein2（ZEB2）in non-small cell lung cancer（NSCLC）and their prognostic significance.Methods Immunohistochemical method was adopted to detect the expressions of E-cadherin and ZEB2 proteins and 144 cases of NSCLC in 52 cases of normal lung tissue more than 5cm away from cancer.Coxregression model was used to analyze the prognostic factors.Results The expression rate of ZEB2 was significantly higher,and the expression of E-cadherin protein was significantly lower in NSCLC tissue（78.5%,34.0%）than those in normal tissue（11.5%,100.0%）（P〈0.05）.The expressions of ZEB2 and E-cadherin proteins were correlated with tumor differentiation degree,TNM stage and lymph node metastasis in NSCLC tissues（P〈0.05）.ZEB2 was negatively correlated with E-cadherin protein in NSCLC tissue（r=-0.444,P=0.000）.Multivariate Coxregression analysis showed ZEB2 and E-cadherin were the independent prognostic factors for NSCLC.Conclusion The upregulated expression of ZEB2 downregulates the expression of E-cadherin protein in NSCLC,which enhances the ability of tumor invasion and metastasis,resulting in a poor prognosis.
