摘  要:目的 探讨转移因子作为免疫增强剂抑制脑干胶质瘤生长的作用.方法 通过立体定向技术制备成年Wistar大鼠C6脑干胶质瘤模型.提取胶质瘤模型大鼠外周血淋巴细胞,制备转移因子.将20只Wistar幼鼠等分为对照组和实验组,在0h分别予以生理盐水和转移因子溶液灌胃.24h后制备幼鼠C6脑干胶质瘤模型.通过MRI检查鉴定胶质瘤模型并测量两组肿瘤体积,采用流式细胞计数法分别检测24 h和96 h两组外周血CD4+和CD8+T淋巴细胞比例.记录两组体征及生存期.结果 幼鼠C6脑干胶质瘤模型制备2周时对照组和实验组肿瘤的平均体积分别为(133.6 ±30.7)mm3和(94.6 ±21.3)mm3,二者差异有统计学意义(P<0.05).24 h对照组和实验组CD4+T细胞比例分别为(10.0±6.5)%和(10.4±5.4)%;CD8+T细胞比例分别为(2.8±1.3)%和(1.9±0.9)%.96 h对照组和实验组CD4+T细胞比例分别为(14.8±1.4)%和(41.3±4.4)%;CD8+T细胞比例分别为(7.5±0.6)%和(24.4±2.8)%.24 h CD4+和CD8+T细胞比例两组差异均无统计学意义(均P>0.05).96 h实验组CD4+和CD8+T细胞比例均高于对照组(均P<0.05).实验组96hCD4+和CD8+T细胞比例均高于24 h(均P<0.05).对照组96 h CD8+T细胞比例高于24 h(P <0.05),24h和96 h的CD4+T细胞比例差异无统计学意义(P>0.05).对照组和实验组平均生存时间分别为(12.8 ±0.6)d和(15.3 ±0.8)d,二者差异有统计学意义(P<0.05).结论 成鼠脑干胶质瘤模型的转移因子可以抑制幼鼠模型肿瘤的生长,从而为脑干胶质瘤的免疫治疗提供了一定的理论依据。Objective To explore whether transfer factors can exert suppressive effect on the growth of brainstem gliomas.Methods C6-Wistar rat brainstem models were used,established by stereotactic implantation of C6 cells into the pons of the rat.Twenty adult rat models were used to extract peripheral blood lymphocytes to prepare transfer factors specific to C6 cells.Twenty juvenile rats were divided into experimental group and control group equally,and were intragastric administered with transfer factors solution and normal saline respectively at the time of 0 hour.Twenty-four hours later rats of the two groups were implanted with C6 cells into its' pons simultaneously.Tumor volumes of the two groups were calculated by diameters measured on T1WI contrast-enhanced MRI image 2 weeks later.The peripheral CD4 + T and CD8 + T lymphocytes were gauged by flow cytometry at the time of 24 h and 96 h.Neurological signs and overall survival were recorded.Results The mean tumor volume of the experimental group (94.6 ± 21.3) mm3 was significantly less than that of the control group (133.6 ± 30.7) mm3,(P 〈 0.05).At the time of 24 h,the proportion of the peripheral CD4 + T lymphocytes was (10.4 ± 5.4) % for the experimental group,and(10.0 ± 6.5) % for the control group;while that of CD8 + T lymphocytes was (1.9 ± 0.9) % and (2.8 ± 1.3) % for the experimental and control group respectively.At the time of 96 h,the proportion of the peripheral CD4 + T lymphocytes was (41.3 ± 4.4) % for the experimental group,and (14.8 ± 1.4) % for the control group;while that of CD8 + T lymphocytes was (24.4 ± 2.8) % and (7.5 ± 0.6) % for the experimental and control group respectively.There was no significant difference of the peripheral CD4 + T and CD8 + T lymphocytes between the two groups at the time of 24 h,While at the 96 h these counts were significantly higher in the experimental group than in the control group (P 〈 0.05).At the time of 96 h,the experimental groups
