脑胶质瘤裸鼠瘤体中胶质瘤干细胞的含量及分布特点

摘　　要：目的 研究脑胶质瘤裸鼠瘤体中胶质瘤干细胞（GSC）的含量及分布特点,并探讨肿瘤大小与GSC含量的关系.方法 胶质瘤干细胞株SU2接种至裸鼠腋部皮下成瘤.按照成瘤后肿瘤长径,分为5 mm组和10 mm两组.利用流式细胞和免疫组化方法,观察并比较两组间GSC的含量和分布.结果 （1）免疫组化显示5 mm组和10 mm组的GSC含量分别为4.02％±1.15％和2.74％±0.51％,可见随着肿瘤的增大而GSC含量减少（t=3.23,P=0.005）;（2）流式细胞检测5 mm组和10 mm组的CD133阳性细胞占全部肿瘤细胞百分比分别为5.03％±2.53％和2.42％±0.79％,5 mm组的CD133阳性细胞含量多于10 mm组（t=3.13,P=0.006）;（3）流式细胞和免疫组化两种方法在检测GSC含量方面无差异性（5 mm组t=1.15,P=0.27;10am组t=1.14,P=0.27）.结论 CD133阳性GSC多位于肿瘤边缘且多分布在较大的血管周围,其含量与肿瘤大小相关,肿瘤直径越小,GSC含量越多。Objective To study the content and distribution characteristics of glioma stem cells （GSC） in human glioma-bearing of nude mice,and to explore the relationship between the content of GSC and tumor size.Methods The situation of tumor-growth were observed after inoculating subcutaneously to nude mice with cells of SU2 GSC lines.According to the tumor diameter,they were divided into two groups,5mm group and 10mm group,the content and distribution of GSC were evaluated by flow cytometric and immunohistochemistry,and all data were analyzed by SPSS 16.0 statistical software as statistically significant difference were considered with P 〈 0.05.Results （1） On immunohistochemistry,the content of GSC in 5mm group and 10mm group were 4.02% ± 1.15% and 2.74% ±0.51%,respectively.The content of GSC were reduced with the increase of the tumors （t =3.23,P =0.005）.（2） On flow cytometric examination,the number of CD133 ＋ cells in 5 mm group and 10 mm group were 5.03% ±2.53 % and 2.42% ± 0.79%,respectively.The content of CD133 ＋ cells in 5mm group were more than that of 10mm group （t =3.13,P =0.006）.（3） There was no difference with the content of GSC between flow cytometry and immunohistochemistry （5 mm group t =1.15,P =0.27,10mm group t =1.14,P =0.27）.Conclusion The GSC was located in the margin of tumor,and nested and gathered around the larger blood vessels.The content of GSC was related with the diameter of tumor,the smaller the diameter,the larger the content.
