摘  要:目的观察内皮抑素(endostatin,ES)对大鼠视网膜新生血管中Flk-1表达的影响。方法实验研究。实验周期为2012年10月至2013年12月。通过缺氧的方法建立大鼠视网膜新生血管模型48只,数字表法随机分为内皮抑索(Es)治疗组及生理盐水(NS)对照组。Es治疗组出氧箱后12、36、72h玻璃体腔内注射ES 1μl,对照组玻璃体腔内注射NS 1μl,并设正常对照组24只。RT-PCR定量检测Flk-1在3组大鼠视网膜中的表达。结果(1)正常大鼠视网膜未见Flk-1表达,缺氧诱导视网膜新生血管后视网膜内Flk-1表达明显增强,Ns对照组及正常对照组比较差异有统计学意义(P〈0.01)。(2)Es治疗组Flk-1表达被显著抑制,与NS对照组比较差异有统计学意义(P〈0.01)。结论(1)缺氧诱导视网膜新生血管后视网膜内Flk-1表达明显增强。(2)ES可有效抑制Flk-1的表达,从而阻断VEGF信号传导途径。Objective To investigate the effects of endostatin on the expression of Flk-1 in the retinal neovascularization. Methods An experimental study. Experimental period were between Janu- ary 2013 and April 2014. Forty-eight mice established the oxygen-induced retinai neovascularization model were randomly divided into NS control group and ES treatment group. ES and normal saline were separately injected into vitreous after 12, 36 and 72 hours out of the oxygen box. At the same time, 24 mice were raised in the room air as the control group. The expression of Flk-1 was exam- ined by RT-PCR method. Results Flk-1 was not expressed in the normal retina. In NS control group, the expression was dramatically increased. There was a significant difference in NS control group and normal control group (P 〈0.01). The expression of Flk-1 was significantly inhibited by ES. There was a significant difference between ES treatment group and NS control group (P 〈0.01). Conclusions After oxygen-induced retinal neovascularization, the expression of Flk-1 is greatly en- hanced. ES can significantly inhibit the expression of Flk-1, and ES can greatly decrease the bioac- tivity of VEGF by inhibiting the expression of Flk-1.
