不同给药时间对姜黄素治疗小鼠溃疡性结肠炎疗效的影响

摘　　要：目的:观察等剂量的姜黄素在不同给药时间内对葡聚糖硫酸钠(DSS)诱导的小鼠溃疡性结肠炎(UC)的治疗作用。方法:将56只雄性BALB/c小鼠随机分为7组(n=8),组1为正常组,组2、组3和组4分别于造模第1天、第5天和第8天开始每日予姜黄素混悬液〔100 mg/(kg?d)〕0.2 m L灌胃,组5、组6和组7分别于造模第1天、第5天和第8天每日予等量0.5%羟甲基纤维素钠(CMC)灌胃,每日记录各组疾病活动指数(DAI)。干预结束后处死小鼠,测量结肠长度,记录远端肠黏膜组织学组织学评分。结果:组2、组3和组4与其疾病对照组相比,DAI和组织学评分降低,结肠长度增长,差异有统计学意义(P<0.05);组3、组4分别与组2的差异有统计学意义(P<0.05),但两者之间差异无统计学意义(P>0.05)。结论:给药时间对姜黄素治疗UC小鼠的效果有明显影响,本实验中姜黄素的干预方式简单易行,疗效确切,为进一步探讨其治疗溃疡性结肠炎的机制提供了有利条件。Objective To observe the therapeutic effects of isodose curcumin on mice with ulcerative colitis induced by dextran sulfate sodium（DSS） at different administration times. Methods 56 male BALB/c mice were randomly divided into 7 groups（n=8）, the Group 1 as normal control, and Group 2, 3 and 4 was given 0.2 ml curcumin suspension with 100 mg/（kg·d） dose by gavage on the 1st, 5th and 8th day, respectively. Group 5, 6 and 7 received equal volume of 0.5 % CMC at the same time point as comparative controls. Disease active indices（DAI） of each group were recorded daily. After intervention mice were sacrifi ced to measure the colon length and score the tissue sections. Results By comparing their disease controls, Group 2, 3 and 4 had lower DAI, decreased histological scores and longer colons, which were statistically signifi cant（P〈0.05）;Group 2 showed signifi cantly different from Group 3 and 4, respectively（P〈0.05）, though no signifi cance was found between the latter two groups（P〉0.05）. Conclusion Administration time obviously influences the therapeutic effects of curcumin on mice with ulcerative colitis, the administrating method of curcumin in this study is simple but effective, which ensures the advantages for further exploration on the potential mechanism of curcumin in treating ulcerative colitis.
