摘  要:目的:通过观察姜黄素治疗后阿尔茨海默病炎症及神经元的变化,初步探讨姜黄素在该疾病中对炎症以及神经元的保护机制。方法:将3月龄APP/V717转基因小鼠及C57BL/6小鼠随机分为C57BL/6野生鼠对照组、姜黄素治疗组(APP/V717转基因小鼠),转基因鼠对照组(APP/V717转基因小鼠)各10只。其中C57BL/6野生鼠对照组和转基因鼠对照组喂养正常的鼠粮,姜黄素治疗组连续喂养含姜黄素的鼠粮9个月(姜黄素按600ppm/d加入到鼠粮中)。喂养后通过水迷宫测试检测小鼠的学习记忆能力;Nissl染色观察小鼠神经细胞的变化;Western blot检测炎症因子IL-1β、TNF-α及凋亡相关的蛋白Bax/Bcl-2的表达;TUNEL法观察小鼠海马区的细胞凋亡情况。结果:1正常鼠粮和含姜黄素鼠粮喂养小鼠体重未发现明显差异;2C57BL/6野生鼠对照组的潜伏期为(21.71±8.69)s,姜黄素治疗组的潜伏期为(25.12±4.28)s,而转基因对照组小鼠的潜伏期为(44.28±8.74)s,姜黄素治疗组小鼠的潜伏期明显低于转基因小鼠,差异有统计学意义(P<0.05);3Nissl染色发现姜黄素治疗组小鼠的神经元细胞与野生型小鼠相似,而转基因小鼠神经元形态不完整,仅有散在的尼氏小体;4IL-1β、TNF-α在转基因小鼠组中表达最高,野生型小鼠中最低,而转基因小鼠在姜黄素治疗后IL-1β、TNF-α的含量明显降低,两两比较差异均有统计学意义(P<0.05);5转基因小鼠的海马区发现较多凋亡的神经元,而在野生型小鼠中几乎未见凋亡神经元,姜黄素治疗组小鼠中见到少量散在的凋亡神经元;6转基因小鼠中Bax的表达明显高于野生型小鼠和姜黄素治疗组小鼠,而Bcl-2的表达量显著低于其余两组,差异有统计学意义(P<0.05)。结论:姜黄素治疗阿尔茨海默病小鼠可以改善小鼠的认知功能,并且有效的减少炎症因子的产生、减少神经元的凋亡。Objective:To explore the changes in neuroinflammation and protection by curcunmin treatment in Alzheimer's disease.Methods:Three months old APP/V717 transgenic mice and C57BL/6mice were randomly divided into 3groups:wild type group(n=10),curcumin treatment group(n=10);transgenic control group(n=10).The wild type group and transgenic control group were fed normal mice food,while curcumin treatment group were fed curcumin food(600ppm/d added to food).Learning and memory ability were detected by morris water maze behavior examination;The change of nerve cells were observed by Nissl staining;the expression of IL-1β,TNF-αand Bax,Bcl-2were detected by western blot;The apoptosis in hippocampus of mice were taken TUNEL assay.Results:1There's no significant difference in normal food fed mice and curcumin fed mice;2The incubation period of wild type group,curcumin treatment group,transgenic control group were(21.71±8.69),(25.12±4.28),and(44.28±8.74)s respectively.The incubation period of curcumin treatment group was significantly lower than that of the transgenic group P〈0.05;3The neurons pattern of curcumin treatment group was similar to that of wild type group,while only scattered Nissl bodies were found in the transgenic control group;4The expression of IL-1β,TNF-αin transgenic control group was the highest,while the lowest in wild type group,the expression of IL-1βand TNF-αin transgenic control group dramatically reduced after curcumin treatment(P〈0.05).The comparison of the differences is statistically significant;5More apoptotic neurons were found in transgenic control group,a few scattered apoptotic neurons were found in curcumin treatment group;6 The expression of Bax in transgenic control group was significantly higher than that of wild type group and curcumin treatment group,while the expression of Bcl-2in transgenic control group was significantly lower than that of wild type group and curcumin treatment group(P〈0.05).Conclusion:Curcumin treatment
