胃癌组织中GOLPH3的表达及临床意义

摘　　要：目的探讨胃癌组织中GOLPH3(Golgi phosphoprotein-3)的表达及临床意义。方法选取148例确诊并接受手术治疗的胃癌患者作为观察组,同时选取20例行胃镜检查的健康成人作为对照组。采用实时荧光定量PCR和免疫组织化学方法对两组组织标本中GOLPH3的mRNA水平和GOLPH3蛋白的表达情况进行检测。根据GOLPH3表达情况将观察组患者分为阳性组和阴性组,分析GOLPH3表达水平与临床病理特征及预后的关系。结果与对照组比较,观察组胃癌组织中GOLPH3 mRNA和蛋白表达水平升高(P<0.05);其中观察组GOLPH3阴性患者46例,阳性患者102例。GOLPH3高水平表达与胃癌组织分型、临床分期、肿瘤浸润程度以及淋巴结转移有密切关系(P<0.05)。GOLPH3阴性患者1、2、3年累积复发率为10.9%、23.9%、30.4%,低于GOLPH3阳性患者的28.4%、38.2%、48.0%(P<0.05);GOLPH阴性患者1、2、3年生存率分别为93.5%、84.8%、69.6%,高于GOLPH3阳性患者的89.2%、70.6%、51.0%(P<0.05)。结论胃癌组织中GOLPH3 mRNA和蛋白异常高表达,与胃癌的病理特征及预后密切相关,可能是胃癌发生、发展的重要机制之一。Objective To investigate the expression of Golgi phosphoprotein-3 （ GOLPH3 ） in gastric cancer and its clinical significance. Methods One-hundred-forty-eight patients with gastric cancer undergoing surgical treatment were selected as study group. Twenty healthy adults receiving gastroscopy were selected as control group. GOLPH3 mRNA levels in two groups were determined by real-time quantitative PCR. GOLPH3 protein levels were analyzed by immunohistoehemistry. The relationship between GOLPH3 protein level and elinicopathological features and prognosis in gastric patients were analyzed. Results Compared with control group, GOLPH3 mRNA and protein levels in study group were significantly increased （ P 〈 0.05 ）. There were 46 cases with negative GOLPH3 and 102 cases with positive GOLPH3. The expression of GOLPH3 was closely correlated with pathological typing, clinical staging, degree of tumor invasion and lymph node metastasis （ P 〈 0.05 ）. The 1-, 2- and 3-year cumulative recurrence rates in GOLPH3 negative patients were lower than those in GOLPH3 positive patients （ 10.9% vs 28.4% ,23.9% vs 38.2% and 30.4% vs 48.0% ,respectively, P 〈 0.05 ）. The 1- ,2- and 3-year cumulative survival rates in GOLPH3 negative patients were higher than those in GOLPH3 positive patients （93.5% vs 89.2% ,84.8% vs 70. 6%, and 69.6% vs 51.0%, respectively, P 〈 0.05 ）. Conclusion GOLPH3 mRNA and protein are highly expressed in gastric cancer tissues, which are closely related to the pathological characteristics and prognosis of gastric cancer.
