摘 要:本研究以双色鸳鸯美人蕉为试材,采用RT-PCR技术扩增查尔酮合成酶基因(CHS)及二氢黄酮醇4-还原酶基因(DFR)序列,并利用RT-q PCR技术检测CHS、DFR及查尔酮异构酶基因(CHI)在双色鸳鸯美人蕉同一朵花不同颜色部位、不同发育时期的表达差异以及在不同花色美人蕉中的表达特性。结果显示:通过扩增分别获得了549 bp的CHS基因片段及570 bp的DFR基因片段,与红掌、百合等相应基因的同源性达70%~95%。基因表达分析表明,CHS及DFR在红色花瓣中的表达量高于黄色花瓣,CHI在黄色花瓣中的表达量高于红色花瓣;CHS与CHI在红色花瓣中随着发育表达量逐渐增大,在黄色花瓣随发育表达量呈先增加后减少的趋势;DFR在红色及黄色花瓣的发育后期表达量逐渐增大;在不同花色美人蕉花瓣中,这三个基因的表达模式各不相同。The red-yellow flower chimera in Canna generalis(Canna generalis Bailey) was used as material and the chalcone synthase gene(Cg CHS) and dihydroflavonol 4-reductase gene(Cg DFR) were cloned using RT-PCR. The549 bp and 570 bp fragment of CHS and DFR genes were cloned, sequence comparison showed that these genes share 70% to 95% identities with each previously reported corresponding genes. The expression patterns of CHS,DFR and CHI(chalcone isomerase) genes were analyzed by Real-time quantitative PCR(RT-q PCR). The results showed that the expression quantity of CHS and DFR genes was higher in red-part than in yellow-part, while CHI gene showed on the contrary. The expression of CHS and CHI genes increased in accordance with flower development in red-part, while in the yellow-part its expression increased in the earlier three developmental stages and then reduced in the fourth stage. The expression pattern of DFR gene was consistent with flower development both in the red- and yellow-parts. The expression pattern of these three genes in Canna generalis varied with different flower color were different with each other.
