摘 要:[目的]研究清肠栓对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)大鼠结肠组织TLR1、TLR2、TLR3及TLR4表达的影响,探讨其免疫调节机制。[方法]将40只SD大鼠随机分为正常组(C组)、模型组(M组)、清肠栓组(QCS组)、美沙拉嗪组(5-ASA组)。造模期间,C组给予蒸馏水,其余组予5%DSS自由饮用6d制备UC模型。药物干预期间,QCS组、5-ASA组分别予相应药物灌肠7d。观察大鼠的一般情况、疾病活动指数(DAI)、结肠指数、结肠组织病理学改变及结肠组织TLR1、TLR2、TLR3及TLR4的表达情况。[结果]1与C组比较,M组DAI评分显著升高(P<0.05);QCS组DAI评分较M组显著下降(P<0.05)。2与C组比较,M组TLR2、TLR4的表达显著升高(P<0.05);与M组比较,QCS组、5-ASA组TLR2、TLR4的表达较M组显著下降(P<0.05)。3各组间的TLR1及TLR3表达差异无统计学意义。[结论]清肠栓对UC有较好的疗效,其作用机制可能是通过下调TLR2、TLR4的表达,影响核因子-κB(NF-κB)的活化,最终减轻机体炎症反应。[Objective]To study the effect of Qingchang suppository on expression of TLR1、TLR2、TLR3and TLR4 in the colon of rats with ulcerative colitis(UC)induced by dextran sulfate sodium(DSS),and discuss its mechanism.[Methods]Forty SD rats were randomly divided into four groups:normal group(C group),model group(M group),Qingchang suppository group(QCS group),mesalazine aminosalicylicacid group(5-ASA group).The normal group were given distilled water,the rats in the other three groups were induced by 5%DSS for 6days for UC model,while the rats in QCS group and 5-ASA group were given the corresponding medicine respectively for 7days.We observed the general condition,disease activity index(DAI),colon index,histopathological changes and expression of TLR1、TLR2、TLR3and TLR4 in the colon.[Results]①Compared with the C group,DAI score was significantly increased in M group(P〈0.05),while that was decreased significantly in QCS group(P〈0.05).②TLR2 and TLR4were expressed higher in M group than the C group(P〈0.05),which were decreased in QCS group and 5-ASA group(P〈0.05).③Expression levels of TLR1 and TLR3in all groups show no significant difference.[Conclusion]Qingchang suppository probably improves the symptoms of UC by down regulating the expression of TLR2、TLR4,then to affect the activation of NF-κB,eventually to relief the inflammatory reaction.
