摘  要:目的 观察前列腺癌(PCa)相关成纤维细胞(CAF)中雄激素受体(AR)对PCa生长能力的作用.方法 用RNA干扰方法降低已经永生化的CAF细胞中AR的表达,得到CAF-ARsi细胞及对照组CAF-sc细胞;用噻唑蓝(MTT)实验、软琼脂糖凝胶实验分别检测CAF中AR表达水平对PCa细胞体外贴壁生长、悬浮生长能力的影响;用实时定量聚合酶链反应(Real-time PCR)检测AR及生长因子的表达水平.结果 成功建立CAF-ARsi细胞及对照组CAF-sc细胞,MTT实验结果显示在第6天时CAF-ARsi细胞生长速度要比CAF-ARsc细胞慢32.40%;软琼脂糖凝胶实验显示PC3与CAF-ARsi共培养时生成的克隆数较CAF-ARsc细胞下降33.47%;而Real-time PCR结果显示干扰CAF细胞中AR表达可明显抑制CAF细胞中胰岛素样生长因子1(IGF-1)、肝细胞生长因子(HGF)等多种生长因子的分泌.结论 CAF细胞中AR通过调控多种生长因子分泌对PCa细胞的生长具有明显的促进作用.Objective To investigate the effect of androgen receptor (AR) in cancer-associated fibroblasts (CAF) on the growth capacity of prostate cancer.Methods We got CAF-ARsi or CAF-sc by knocking down the AR in CAF which has already been immortalized by small interfering RNA (siRNA).The methyl thiazol tetrazolium (MTT) and soft agar assays were performed to study the PC3 adherent growth ability and suspended growth ability.The expression levels of AR and growth factors in CAFs were detected by the real-time quantitative polymerase chain reaction (Real-time PCR).Results We cultured and immortalized the resultant cells successfully.The MTF showed that the growth of PC3 cells cocultured with CAF-ARsi was reduced by 32.40% as compared with the control cells.The soft agar assay indicated that the colonies of PC3 cells cocultured with CAF-ARsi were decreased by 33.47% as compared with those cocultured with CAF-sc 33.47%.Real-time PCR revealed that the expression levels of insulin-like growth factors-1 (IGF-1),fibroblast growth factor-7 (FGF-7),FGF-10,stromal cell derived factor-1 (SDF-1),hepatocyte growth factor (HGF),and transforming growth factor (TGF)-β2 were decreased significantly in the CAF-ARsi cells.Conclusion The AR in CAFs could promote the growth capacity of prostate cancer.
