摘 要:目的:建立超高效液相色谱串联质谱法(UPLC-MS/MS法)同时测定白芍饮片中10种主要成分(没食子酸、原儿茶酸、芍药苷亚硫酸酯、氧化芍药苷、儿茶素、没食子酸甲酯、芍药内酯苷、芍药苷、五没食子酰葡萄糖、苯甲酰芍药苷)的含量。方法:采用UPLC-MS/MS法,负离子多反应监测(MRM)模式进行含量测定。色谱条件:采用Waters CORTECS C18(2.1 mm×100 mm,1.6μm)色谱柱,以乙腈(A)-0.1%甲酸水(B)为流动相,梯度洗脱,流速0.25 m L·min-1,柱温45℃。结果:在所设定的色谱条件下,5 min内定量分析白芍中10种主要成分在考察的浓度范围内呈良好的线性关系(r〉0.996 6);回收率和RSD分别在97.3%~102.0%和3.1%~5.1%范围内。结论:本研究所建立的同时测定白芍中10种主要成分(没食子酸,原儿茶酸,芍药苷亚硫酸酯,氧化芍药苷,儿茶素,没食子酸甲酯,芍药内酯苷,芍药苷,五没食子酰葡萄糖,苯甲酰芍药苷)的UPLC-MS/MS定量分析方法简便、快捷、准确,为白芍饮片质量控制提供新的技术手段。Objective: To establish a rapid,accurate UPLC- MS / MS method for the determination of 10 major components( gallic acid,potocatechuic acid,paeoniflorin sulfonate,oxypaeoniflorin,catechin,methyl gallate,albiflorin,paeoniflorin,pentagalloylglucose,benzoylpaeoniflorin) in Radix Paeoniae Alba pieces. Methods: The UPLC- MS /MS assay was performed on a Waters CORTECS C18( 2. 1 mm × 100 mm,1. 6 μm) column with acetonitrile- water( containing 0. 1% formic acid) as the mobile phase by gradient elution at a flow rate of 0. 25 m L·min- 1. The column temperature was set at 45 ℃. MS detection was performed with multiple reaction monitoring( MRM) mode using negative electrospray ionization. Results: Under the optimized chromatographic conditions,good separation of 10 target components were obtained within 5 min. Satisfactory linearity was achieved within the investigated linear range and with fine determination coefficient( r 0. 996 6),the overall recoveries were ranged from 97. 3%- 102. 0% with the RSD ranging from 3. 1%- 5. 1%. Conclusion: It is the first report of simultaneous analysis of 10 major components( gallic acid,potocatechuic acid,paeoniflorin sulfonate,oxypaeoniflorin,catechin,methyl gallate,albiflorin,paeoniflorin,pentagalloylglucose,benzoylpaeoniflorin) in Radix Paeoniae Alba by using UPLC- MS / MS method,which affords a highly sensitive,specific,speedy and efficient method for quality control of Radix Paeoniae Alba pieces.
