摘 要:目的分析中国北方汉族人群21个常染色体STR基因座和1个Y染色体STR基因座的遗传多态性,为个体识别和亲权鉴定提供遗传学数据。方法用GlobalFiler PCR AmplificationKit荧光标记试剂盒对1081例中国北方汉族无关个体的DNA进行PCR扩增.3500XL遗传分析仪电泳分析,用GeneMapperID-X软件分析等位基因片段大小,用Modified-Powerstates.xls分析软件进行等位基因频率和法医学常用参数统计分析。结果在中国北方汉族人群中.21个常染色体STR基因座的遗传多态性高,杂合度(H)分布在0.604~0.939之间,随机匹配率(MP)分布在0.007~0.206之间,个体识别力(PD)在0.794~0.993之间,非父排除率(PE)在0.296~0.875之间,典型父权指数(TPI)在1.26-8.19之间,多态性息含量(PIC)在0.55.0.94之间:DYS391基因座共检出6个等位基因,GD值为0.4158。结论中国北方汉族人群21个常染色体STR基因座具有较高多态性,可以用于法医学亲权鉴定和个体识别,也可以用于人类学和遗传学研究。1个Y染色体STR基因座和1个Y-InDel遗传标记可以辅助判断被鉴定人性别。Objective To analyze the genetic polymorphism of twenty-one autosomal STR loci and one Y chromosomal STR locus in Chinese Northern Han population and to provide genetic data for the forensic application of individual and paternity identification. Method The blood samples of 1081 unrelated individuals from Chinese Northern Han population were collected. GlobalFilerg PCR Amplification Kit with fluorescent labeling was used for PCR amplification. 3500XL Genetic Analyzer was used to perform electrophoresis analysis. GeneMapper~ ID-X software was used to analyze the allele sizes, and Modified- Powerstates spreadsheet was used to count allele frequencies and other forensic parameters. Results The twenty-one autosomal STR loci were highly polymorphic in Chinese Northern Han population. Heterozygosity (H) ranged from 0.604 to 0.939. Match Probability (MP) varied from 0.007 to 0.206. Power of Discrimination (PD) ranged from 0.794 to 0.993. Probability of Exclusion (PE) ranged from 0.296 to 0.875. Typical Paternity Index (TPI) ranged from 1.26 to 8.19. Polymorphism Information Content (PIC) was from 0.55 to 0.94. Six alleles were detected in DYS391 locus and Genetic Diversity (GD) was 0.4158. Conclusion The twenty-one autosomal STR loci have relatively high polymorphism in Chinese Northern Han population, which could be used for population genetics and forensic studies. One Y chromosomal STR and one Y-InDel genetic marker could be employed for gender determination.
