摘  要:[目的]筛选并鉴定生物表面活性剂,同时分析其目标产物结构。[方法]运用油平板法、排油圈法等方法对从葡萄中筛选出的72株菌株进行初筛与复筛,初步筛选出产生物表面活性剂的最佳菌株,对其进行菌种鉴定,并通过薄层层析试验、红外光谱分析等鉴定生物表面活性剂类型。[结果]从72株葡萄内生菌中分离筛选得到9株产生物表面活性剂的菌株,其中菌株C2J6发酵液排油圈为最大,通过形态特征、生理生化试验及26S r DNA序列分析,初步鉴定该菌为黑曲霉(Aspergillus niger)。通过薄层色谱和红外光谱分析表明,菌株C2J6在代谢过程中能产生脂肽类表面活性物质。[结论]试验可为研究脂肽类生物表面活性剂的产生与应用打下基础。[ Objective ] To screen and identify biosurfactants, analyze structure of target products. [ Method] Primary screening and rescreening of 72 strains were conducted by using oil plate method, oil displacement method, the optimal strain was primarily screened out and identified, thin layer chromatography analysis test, infrared spectrum were conducted to analyze biosurfactants. [ Result ] Separating 9 out of 72 grapes endophyte strains screened surface active agent by the screening plate method, and drain ring rescreening method. Strain broth which named C2J6 oil drain ring maximized. According to morphological, physiological and biochemical tests and sequence analysis that 26sRNA preliminarily is identified as Aspergillus niger. Thin layer chromatography and infrared spectroscopy showed that strain C2J6 in the metabolic process produces lipopeptide surfactant. [ Conclusion] The study can lay a foundation for production and application of lipopeptide biosurfactants.
