摘  要:目的探讨依托咪酯对急性脑缺血大鼠海马CA1区细胞凋亡与胱天蛋白酶(caspase)-3及胱天蛋白酶(caspase)-10表达的影响。方法线栓法制作大鼠大脑中动脉缺血(MCAO)再灌注模型。应用原位末端标记法检测细胞凋亡,蛋白印迹法检测caspase-3及caspase-10蛋白表达。结果 1脑缺血组大鼠海马CA1区3h即出现凋亡阳性细胞,48h达到高峰,72h凋亡阳性细胞的数量开始下降。依托咪酯组大鼠海马CA1区凋亡细胞数于给药后再灌注6~72h各时间点均低于脑缺血组(P〈0.05)。2依托咪酯组大鼠海马CAl区caspase-3及caspase-10蛋白表达于给药后再灌注6--72h各时间点均低于脑缺血组(P〈0.05)。结论依托咪酯能够抑制急性脑缺血损伤引起的细胞凋亡。Objective To investigate the effect of etomidate on acute cerebral ischemia in rat hippocampal CA1 cells apoptosis and the caspase-3and caspase-10 expression influence.Methods With intraluminal suture method in rats with middle cerebral artery ischemia reperfusion model(MCAO).Application of in situend labeling method to detect apoptosis,Western-blotting assay for detection of caspase-3and caspase-10 protein expression.Results 1Cerebral ischemia rats hippocampal area CA1 is of 3happearance of apoptosis positive cells,48 h peak,72 hapoptosis positive cells began to decline.Etomidate group hippocampal CA1 area of rats after administration of the number of apoptotic cells in reperfusion 6-72 hat each time point were lower than those in ischemia group(P〈0.05).2The etomidate group in hippocampal CA1 region of rats with caspase-3and caspase-10 protein expression in after the administration of 6-72 hreperfusion time at each time point were lower than those in ischemia group(P0.05).Conclusion Etomidate can inhibit acute cerebral ischemia induced cell apoptosis.
